Dermatófitos são fungos que causam uma variedade de micoses de pele, cabelos, pelos e unhas em animais e humanos. Eles acometem tecidos queratinizados provocando dermatofitoses, como a causada pelo patógeno humano Trichophyton rubrum. O tratamento da infecção fúngica é desafiador, pois o fungo pode apresentar resistência aos antifúngicos. As bombas de efluxo, presentes na membrana celular, fazem o efluxo de diferentes antifúngicos, sendo um mecanismo frequentemente associado à resistência a múltiplas drogas (MDR - multidrug resistance). Além disso, os fungos podem realizar ajustes finos via splicing alternativo (SA) como estratégia adaptativa frente a diferentes estímulos. A retenção de íntron (RI) é um tipo de SA, e parece ser o mais frequente em Ascomycetos. Dessa forma, o objetivo desse projeto foi investigar a ocorrência de SA em genes que codificam para proteínas transportadoras MDRs em T. rubrum após exposição a drogas antifúngicas. Dados de RNA-Seq de T. rubrum em resposta aos antifúngicos ácido undecanóico e acriflavina foram utilizados para a triagem destes genes. O gene identificado como TERG_04309 que codifica um transportador ABC se destacou por apresentar um número expressivo de RI 3 e 4. Subsequentemente, foram realizados cultivos de T. rubrum na presença dos antifúngicos Ácido Undecanóico (UDA), Caspofungina (CASP), Itraconazol (ITR) e Terbinafina (TRB) e a modulação das isoformas da TERG_04309 foi avaliada por RT-PCR e RT-qPCR, com o objetivo de verificar se este fenômeno se aplicaria a outros fármacos de diferentes mecanismos de ação. Resultados de RT-qPCR demonstraram a indução da isoforma com o íntron-3 retido após 12 h de exposição ao ITR e a indução da isoforma com íntron-4 retido, após 1h e 12 h de exposição a CASP. Foi observado também a indução proeminente da isoforma convencional após exposição do fungo a TRB e ITR. Paralelamente, genes que putativamente interagem com TERG_04309 tiveram suas modulações avaliadas. Um deles, a TERG_04224, que também codifica uma bomba de efluxo teve sua expressão induzida nas condições de 1h CASP e 12h ITR. Isto indica a co-expressão de TERG_04309 e TERG_04224. Em muitos casos, a RI desencadeia proteínas truncadas e com perdas de domínios, assim seria concebível que as isoformas provenientes de SA fossem não funcionais. Entretanto, o alinhamento das isoformas alternativas com proteínas ortólogas mostra semelhanças entre elas, o que sugere que as isoformas oriundas da RI do gene TERG_04309 podem manter suas funções, com prováveis alterações na afinidade e efetividade na extrusão. Adicionalmente, a modulação das isoformas de TERG_04309 foi avaliada em biofilmes maduros de T. rubrum após exposição às drogas CASP e TRB por 1h. Foi evidenciada a indução de ambas as isoformas alternativas após a exposição à CASP, sugerindo que elas têm função no efluxo de drogas. Portanto, os presentes resultados indicam que o T. rubrum tem respostas tempo e composto-específicos, que envolve além dos mecanismos regulatórios convencionais, um mecanismo alternativo proveniente de SA, que ocorre tanto nas fases de crescimento planctônico como na forma de biofilmes.

Dermatophytes are fungi that cause various mycoses of skin, hair, and nails in animals and humans. They affect keratinized tissues causing dermatophytoses, such as that caused by the human pathogen Trichophyton rubrum. The treatment of fungal infection is challenging, as the fungus can show resistance to antifungals. Efflux pumps, present in the cell membrane, extrude different antifungals from the cells, being a mechanism often associated with multidrug resistance (MDR). In addition, fungi can make fine adjustments via alternative splicing (SA) as an adaptive strategy in the face of different stimuli. Intron retention (IR) is one of the types of AS and appears to be the most frequent in Ascomycetes. Thus, this project's objective was to investigate the occurrence of AS in genes that code for multidrug transporter proteins in T. rubrum after exposure to antifungal drugs. We used T. rubrum RNA-Seq data in response to undecanoic acid and acriflavine antifungals to screen for these genes. The gene identified by TERG_04309, which encodes a multidrug transporter protein, stood out for having an expressive number of introns 3 and 4 retentions. Subsequently, cultures of T. rubrum were performed in the presence of the antifungals Undecanoic Acid (UDA), Caspofungin (CASP), Itraconazole (ITR), and Terbinafine (TRB), and the modulation of the TERG_04309 isoforms was evaluated by RT-PCR and RT-qPCR. The objective was to assess whether this phenomenon would apply to other drugs with different mechanisms of action. Results from RT-qPCR demonstrated that the induction of the isoform with retained intron 3 was detected after 1 h of exposure to ITR and induction of the isoform with retained intron-4 after 1 h and 12 h of exposure to CASP. It was also observed a prominent induction of the conventional isoform after exposure of the fungus to TRB and ITR. In parallel, genes that putatively interact with TERG_04309 had their modulations evaluated. One of them, TERG_04224, which also encodes an efflux pump, had its expression induced under CASP conditions at 1h and ITR conditions at 12h. This result indicates co-expression of TERG_04309 and TERG_04224. It's well known in many cases that IR triggers truncated proteins with a loss of domains, so it is conceivable that alternative isoforms would be non-functional. However, the alignment of alternative isoforms with orthologues displayed similarities between them, which suggested that isoforms obtained from IR of gene identified by TERG_04309 might keep their functions, with possible changes regarding affinity or effectivity in extrusion. In addition, the modulation of the TERG_04309 isoforms was evaluated in mature T. rubrum biofilms after exposure to CASP and TRB drugs for 1 h. The induction of both alternative isoforms was evidenced after exposure to CASP, suggesting that they have a role in the efflux of drugs. Therefore, the present results indicate that T. rubrum has time-and compound-specific responses, which involves, in addition to conventional regulatory mechanisms, an alternative mechanism originating from AS, which occurs both in planktonic growth phases and in the form of biofilms.
