O sistema purinérgico consiste em duas grandes famílias de receptores do tipo P2 - P2X e P2Y. Ambos são ativados pelo trifosfato de adenosina (ATP) e estão presentes em várias estruturas encefálicas, incluindo aquelas envolvidas com o controle emocional e processos de memória. Evidências sugerem que estes receptores podem modular respostas de medo, porém pouco se sabe sobre seu papel em situações aversivas aprendidas. Portanto, este estudo teve por objetivo investigar o envolvimento de receptores purinérgicos na expressão e extinção da memória de medo condicionado. Foram utilizados camundongos do tipo selvagem (WT) e knockout para o receptor P2X7 (P2X7-/-). No protocolo de Medo Condicionado ao Contexto (MCC), os animais WT tratados via intraperitoneal (i.p.) 10 min antes da sessão de extinção com a maior dose do antagonista não seletivo de receptores P2 (P178) exibiram um aumento na expressão da resposta condicionada e prejuízo na retenção da memória de extinção. Resultados similares foram obtidos com o antagonista seletivo de receptores do subtipo P2X7 (A438079), mas não com o antagonista seletivo de receptores do subtipo P2Y1 (MRS2179). Contudo, tais respostas não foram observadas quando a administração do antagonista não seletivo de receptores P2 (P178) foi realizada imediatamente após a sessão de extinção. No paradigma do Medo Condicionado ao Som (MCS), animais WT tratados 10 min antes da sessão de extinção com antagonista seletivo de receptores P2X7 (A438079) apresentaram resistência no aprendizado da extinção, prejuízo na retenção dessa memória e generalização do medo. Nenhum efeito foi observado quando a droga foi administrada imediatamente após a sessão de extinção. Os animais P2X7-/- exibiram perfil comportamental semelhante ao tratamento farmacológico em ambos os testes comportamentais. Os resultados sugerem que os receptores P2X7 estão envolvidos com resposta aversiva e o aprendizado associado à extinção da memória aversiva. Assim, o sistema purinérgico é um possível candidato para o desenvolvimento de novas terapias para o tratamento de transtornos de ansiedade.

The purinergic system consists of two families of P2 receptors - P2X and P2Y. Both are activated by adenosine triphosphate (ATP) and are present in several brain regions, including those involved in emotional control behavior and memory processes. Hence, they seem to modulate fear response, however, few is known about their role in fear learning memory. Therefore, this study investigated the involvement of purinergic receptors in the expression and extinction of aversive memory. Wild-type (WT) and P2X7 receptor knockout (P2X7-/-) mice were used. In the Contextual Fear Conditioning protocol, WT mice received intraperitoneal (i.p.) injection 10 min prior the extinction session with the highest dose of the nonselective P2R antagonist (P178) exhibited an increase in the fear expression response and impaired retention of extinction memory. Similar results were observed with the selective P2X7R antagonist (A438079), but not with the selective P2Y1R antagonist (MRS2179). However, such responses were not observed when the administration of the non-selective P2 receptor antagonist (P178) was performed immediately after the extinction session. In the Cued Fear Conditioning protocol, WT mice treated 10 min before the extinction session with the selective P2X7R antagonist (A438079) showed resistance in the extinction learning, impaired retention of extinction memory and fear generalization. No effect was observed when the drug was administered immediately after the extinction session. The P2X7-/- mice showed behavior profile similar to the pharmacological treatment in both behavior protocols. These results suggest that P2X7 receptors are involved with aversive response and learning associated with the extinction of aversive memory. Thus, the purinergic system is a possible candidate for the development of new therapies for the treatment of anxiety disorders.
