O hipocampo é uma estrutura bilateral que, em roedores, se estende do eixo dorsal ao ventral, podendo ser divido em hipocampo dorsal (HD) e hipocampo ventral (HV). Apesar de ser conhecido principalmente por sua participação na formação de memória e na navegação espacial, o hipocampo ocupa também um papel fundamental na ansiedade. Sabe-se que a neurotransmissão serotonérgica, principalmente a mediada por receptores 5-HT1A, desempenha um papel crítico na modulação de comportamentos relacionados à ansiedade e que o aumento desta neurotransmissão no hipocampo é requerida para a eficácia de drogas utilizadas no tratamento dos transtornos de ansiedade, tais como a imipramina e a fluoxetina. A distribuição dos receptores 5-HT1A e sua ação no hipocampo é diferente ao longo do eixo dorsoventral. Mais especificamente, no HD, a ativação dos receptores 5-HT1A causa um efeito do tipo ansiogênico em testes comportamentais. Por outro lado, no HV, apesar de escassa a literatura, a ativação destes receptores parece ser ansiolítica. Portanto, o objetivo do presente estudo foi estender as investigações acerca da dualidade do efeito da ativação dos receptores 5- HT1A no HD e no HV do hipocampo e entender os efeitos ocasionados nestes receptores pelo tratamento crônico com antidepressivos. Inicialmente, avaliamos as consequências de injeções intra-HD e intra-HV de serotonina ou do agonista de 5-HT1A 8-OH-DPAT em ratos submetidos ao teste de beber punido de Vogel. Também no HV, foram analisados os efeitos decorrentes da injeção do antagonista de receptores 5-HT1A WAY-100635 e as consequências da combinação das injeções locais deste antagonista com a de serotonina e 8-OH-DPAT. Na segunda etapa, avaliamos se o tratamento crônico com a fluoxetina e a imipramina, altera a resposta modulada pela injeção intra-HD de 8-OH-DPAT. Os resultados mostram que a injeção dos agonistas intra-HD e intra-HV provocaram, respectivamente, respostas do tipo ansiogênica e ansiolítica no teste de beber punido de Vogel. A injeção de WAY100635 no HV provocou resposta do tipo ansiogênica e a dose subefetiva deste antagonista foi capaz de bloquear o efeito ansiolítico da injeção de serotonina e de 8-OH-DPAT no HV. Por fim, foi possível observarmos que o tratamento crônico com imipramina ou fluoxetina foi capaz de diminuir o efeito ansiogênico causado pela administração de 8-OH-DPAT no HD. Estes resultados sugerem que os receptores 5-HT1A no HD e no HV desempenham papéis opostos no processamento da ansiedade, e que o tratamento crônico com antidepressivos diminui a resposta modulada por estes receptores no HD.

The hippocampus is a bilateral structure that, in rodents, extends from the dorsal to the ventral axis, and can be divided into dorsal hippocampus (DH) and ventral hippocampus (VH). Despite being known mainly for its participation in memory formation and spatial navigation, the hippocampus also plays a fundamental role in anxiety. It is known that serotonergic neurotransmission, mainly mediated by 5-HT1A receptors, plays a critical role in modulating anxiety-related behaviors and that the increase in this neurotransmission in the hippocampus is required for the effectiveness of drugs used to treat anxiety disorder, such as imipramine and fluoxetine. The distribution of 5-HT1A receptors and their action in the hippocampus is different along the dorsoventral axis. More specifically, in DH, the activation of 5-HT1A receptors causes an anxiogenic like effect in behavioral tests. On the other hand, in VH, despite the scarce literature, the activation of these receptors seems to be anxiolytic. Therefore, the aim of the present study was to extend the investigations about the dual effect of the activation of 5-HT1A receptors in the DH and VH and to understand the effects caused in these receptors by chronic treatment with antidepressants. Initially, we measured the consequences of intra-DH and intra-VH injections of serotonin or the 5-HT1A 8-OH-DPAT agonist in rats subjected to the Vogel punished test. Also in VH, we analyzed the effects resulting from the injection of the 5-HT1A receptor antagonist WAY-100635 and the consequences of combining local injections of this antagonist with serotonin and 8-OH-DPAT. In the second step, we evaluated whether chronic treatment with fluoxetine and imipramine alters the response modulated by the intraDH injection of 8-OH-DPAT. The results show that the injection of intra-DH and intra-VH agonists provoked, respectively, anxiogenic and anxiolytic responses in the Vogel punished test. The injection of WAY-100635 in VH caused an anxiogenic response and the subefective dose of this antagonist was able to block the anxiolytic effect of the injection of serotonin and 8-OH-DPAT in VH. Finally, it was possible to observe that chronic treatment with imipramine or fluoxetine was able to decrease the anxiogenic effect caused by the administration of 8-OHDPAT in DH. These results suggest that 5-HT1A receptors in DH and VH play opposite roles in the processing of anxiety, and that chronic treatment with antidepressants decreases the response modulated by these receptors in HD.
