O milho geneticamente modificado, com expressão de toxinas inseticidas derivadas de genes da bactéria entomopatogênica Bacillus thuringiensis Berliner (Bt), atualmente ocupa em quase sua totalidade a área destinada ao cultivo do cereal no Brasil. O principal alvo desta ferramenta é a lagarta-do-cartucho Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae), praga de maior relevância para a cultura do milho no país. A durabilidade das tecnologias Bt é grandemente ameaçada pela evolução da resistência desta praga-alvo devido a uma combinação de fatores, tais como: maciça adoção de milho Bt, características bioecológicas da espécie S. frugiperda, pressão de seleção constante sobre a praga, plantio insuficiente de áreas refúgio, carência de estratégias viáveis e eficazes dentro do Manejo da Resistência de Insetos (MRI). Casos de redução crítica da eficácia de algumas toxinas Bt do grupo Cry1 no controle de S. frugiperda foram documentados no Brasil. A toxina inseticida Vip3Aa20 é a mais recente tecnologia comercialmente disponível no âmbito dos cultivos Bt. A longevidade da eficácia de Vip3Aa20 no controle de S. frugiperda pode ser ampliada com a implantação da estratégia de MRI conhecida como alta dose/refúgio. Áreas refúgio são fonte de indivíduos de genótipo suscetível (ss); entretanto, a existência de indivíduos de genótipo heterozigoto (rs), carreadores de alelos da resistência, deve ser levada em conta na estruturação de um refúgio eficaz. A manutenção da suscetibilidade da população da praga-alvo é positivamente influenciada quando o valor adaptativo de indivíduos carreadores de alelos r é inferior ao valor adaptativo dos indivíduos suscetíveis. A ocorrência e intensidade do custo adaptativo associado à resistência é uma das determinantes na evolução da resistência. Programar áreas refúgio visando maximizar o custo adaptativo dos carreadores de alelos r tem grande valor no contexto do MRI. A possibilidade de uso do inseticida químico Fortenza 600® no tratamento de sementes (TS) de milho não-Bt utilizado para formar áreas refúgio pode ser uma ferramenta eficaz para ampliar a magnitude e/ou a dominância do custo adaptativo. Para tal investigação, amostras de S. frugiperda foram coletadas em quatro regiões produtoras de milho no Brasil. A técnica F2 Screen foi utilizada para estimar a frequência de alelos da resistência à Vip3Aa20 dos locais amostrados e selecionar, em laboratório, uma linhagem resistente. A linhagem selecionada, LRV- MT Vip-R, teve sua resistência fenotípica demonstrada em bioensaio, com sobrevivência larval e viabilidade de ovos sem diferenças significativas ao se alimentar de tecido fresco de milho não-Bt e de milho VipteraTM. Os estudos indicaram possível custo adaptativo na sobrevivência larval e viabilidade de adultos na linhagem LRV-MT Vip-R. A linhagem heterozigótica LRV-MT♀ x SUS♂ (RS-1) apresentou valor adaptativo semelhante à linhagem SUS. A linhagem heterozigótica LRV- MT♂ x SUS♀ (RS-2), em diversos parâmetros biológicos, apresentou tendência de maior valor adaptativo em relação às outras linhagens estudadas. A oferta de tecido foliar de milho não-Bt com idade de 21 DAE, oriundo de sementes tratadas com Fortenza 600®, causou redução na viabilidade de adultos da linhagem suscetível e linhagens heterozigóticas; a viabilidade de adultos da linhagem LRV-MT Vip-R não foi afetada. Os parâmetros de fecundidade total fêmea-1 e longevidade de fêmeas das quatro linhagens não diferiram ao se alimentarem de tecido tratado em comparação ao tecido sem tratamento. A linhagem suscetível apresentou considerável número de adultos com alterações morfológicas visuais quando alimentada com tecido tratado. A utilização de Fortenza 600® em TS de milho para implantação de áreas refúgio não se mostrou eficaz em potencializar o custo adaptativo associado à resistência de S. frugiperda à Vip3Aa20.

Genetically modified maize expressing insecticidal toxins originated from Bacillus thuringiensis Berliner (Bt) entomopathogenic bacteria genes, currently occupies almost entirely the area intended to the cereal growth in Brazil. The main target of this tool is Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae), most relevant pest for maize production in the country. The Bt technologies durability is largely threatened by the target-pest resistance evolution due to a combination of factors such as the massive adoption of Bt maize, bio-ecological characteristics of S. frugiperda, constant selection pressure over the pest, insufficient refuge areas, the lack of practicable and efficient strategies in the Insect Resistance Management (IRM). Critical reduction in effectiveness of some Bt toxins from Cry1 group in the control of S. frugiperda was documented in Brazil. Vip3Aa20 insecticidal toxin is the latest technology commercially available in the contest of Bt plants. The longevity of Vip3Aa20 efficiency in S. frugiperda control can be increased by the implementation of the strategy of MIR known as high-dose/refuge. Refuge areas are source of susceptible phenotype individuals (ss) to the Bt toxins; however, the existence of heterozygous phenotype individuals (rs), resistance allele carriers, must be considered when structuring an effective refuge. The maintenance of target-pest population susceptibility is positively influenced when the fitness value of r allele carriers is lower than the fitness value of the susceptible individuals. The occurrence and intensity of fitness cost associated with resistance are one of the resistance evolution determinants. Planning refuge areas in order to maximize fitness cost of r alleles carriers has great value in the IRM context. The use of chemical insecticide Fortenza 600® in seed treatment of non-Bt maize, used to grow refuge areas, can be a helpful tool to extend the magnitude and/or dominance of fitness cost. S. frugiperda was sampled in four maize growing regions in Brazil. F2 screen technique was used to estimate frequency of resistance alleles to Vip3Aa20 in the sampled areas and to select, in laboratory, a resistant strain. The phenotypic resistance of LRV-MT Vip-R was demonstrated in a bioassay, with no significant difference of larval survival and eggs viability when fed with non-Bt maize and VipteraTM maize fresh leave tissue. The tests showed the possibility of fitness cost presence in LRV-MT Vip-R larval survival and adult viability. LRV-MT♀ x SUS♂ (RS-1) showed similar fitness value to the suscetible strain. For several biological parameters, LRV-MT♂ x SUS♀ (RS-2) presented higher adaptative value comparing to the other investigated strains. Adult viability of SUS and heterozigous strains was decreased when larvae were fed with Non-Bt maize leaf tissue aged 21 DAE, with Fortenza 600FS®; the adult viability of the resistance strain did not show significant difference. Total fertility female-1 and female longevity of the four strains did not differ when fed with treated fresh tissue comparing to non-treated. The susceptible strain showed high number of adults with morphological deformations when the strain was fed with treated fresh foliar tissue. The adoption of Fortenza 600FS® in maize seed treatment for the implementation of refuge is not an efficient tool to increase fitness cost associated with S. frugiperda resistance to Vip3Aa20.
