As lentilhas dágua (Lemnaceae), também conhecidas como "duckweeds", são pequenas monocotiledoneas aquáticas que se dividem em duas subfamilias: Lemnoideae e Wolffioideae. Essas espécies possuem diversas cararcteristicas de interesse agronômico, aquanômico e ecológico sendo as principais delas: rápida multiplicação de suas pequenas folhas, denominadas frondes; seu alto valor nutricional e proteico; sua capacidade de remoção de resíduos e fitorremediação do local onde habitam; suas aplicações farmacológicas e biotecnológicas; e a possibilidade de produção em larga escala em biorreatores. Devido às suas qualidades, essas plantas vem sendo estudadas para serem introduzidas em protocolos de cultura de tecidos, permitindo assim a manipulação genética. O presente trabalho teve como objetivo estabelecer protocolos de cultura de tecidos para três genótipos de Lemnoideae: 8627 e DWC124, da espécie Lemna minor, e 7498 da espécie Spirodela polyrhiza. Neste estudo, foram conduzidos experimentos para obtenção de indução de calos, regeneração de plantas, testes de resistência de frondes a antibiótico, regeneração de plantas a partir de calos na presença de antibióticos e também a transformação genética de calos do genótipo 8627 de Lemna minor via Agrobacterium tumefaciens com o vetor pCAMBIA2301. Este vetor expressa o genes uidA que codifica a proteína β-glucuronidase - GUS e o nptII que confere resistência à canamicina. O tratamento que melhor permitiu indução de calos para os três genótipos estudados, foi o com adição de 0,25 mg.L-1 de 2,4-D e 1 mg.L-1 de TDZ ao meio de cultivo MS na condição de escuro e a partir de frondes inteiras (intactas). Para regeneração de plantas, o meio de cultivo MS diluído, e o tratamento com adição de 0,2 mg.L-1 de BAP, foram os mais eficientes em promover a regeneração de novas folhas a partir dos calos. Para o genótipo 8627 de L. minor, calos com idade entre 120 e 130 dias, ou entre 140 e 150 dias, foram os que apresentaram maior taxa de regeneração, de 88%. Para o genótipo DWC124 de L. minor, calos com idade entre 120 e 130 dias promoveram um a taxa de 74% de regeneração. E os calos do genótipo 7498 de S. polyrhiza, apresentaram 44% de regeneração a partir de calos com idades entre 45 e 55 dias ou entre 85 e 95 dias. A partir dos resultados dos experimentos de resistência a antibióticos, onde se foram testadas diferentes concentraçoes de canamicina, espectinomicina, estreptomicina, geneticina, tobramicina e timentin, pode-se definir quais concentrações se mostraram letais para cada um dos três genótipos e quais permitiram o crescimento de cerca de 50% das plantas, concentração ideal para ser utilizada em seleção de plantas em experimentos de transformação. A transformação genética de calos do genótipo 8627 de L. minor resultou na eficiência de 82,2 % de expressão transiente do GUS em calos e somente 2,7% nas frondes. Pode-se então induzir calos nos três genótipos, regenerar plantas a partir desses, e transformar de maneira transiente calos de L. minor 8627.

The duckweed family (Lemnacceae) are small aquatic monocots that are divided in two subfamilys: Lemnoideae e Wolffioideae. These species have several characteristics of agronomical, aquanomic and ecological interest, the main ones are: rapid multiplication of their small leaves, called fronds; their high nutritional and protein value; their capacity to remove residues and to phytorremediate their habitat; their pharmacological and biotechnological applications; and the possibility of a large-scale production in bioreactors. Due to their qualities, these plants have been studied to be introduced in tissue culture protocols, thus allowing their genetic manipulation. This work aimed to establish tissue culture protocols for three genotypes of Lemnoideae: 8627 and DWC124 of the species Lemna minor and 7498 of the species Spirodela polyrhiza. In this study, experiments were conducted to obtain callus induction, plant regeneration, tests of fronds resistance and growth in antibiotic, plant regeneration from calli in the presence of antibiotics, and genetic transformation of L. minor 8627 genotype via Agrobacterium tumefaciens harboring the pCAMBIA2301 vector. This vector express the uidA gene that codifies the enzyme β-glucuronidase GUS and the nptII gene that confers resistance to kanamicin. The treatment that best allowed callus induction for the three genotypes studied, was the one with 0,25 mg.L-1 of 2,4- D and 1 mg.L-1 of TDZ added to the MS culture media in the dark condition using entire (intact) fronds. For plant regeneration, the diluted MS culture medium and the treatment with addition of 0.2 mg.L-1 of BAP were the most efficient in promoting the regeneration of new leaves from the calli. For L. minor 8627, calli with ages between 120 and 130 days, or between 140 and 150 days, were the ones with the highest regeneration rate, 88%. For L. minor DWC124, calli with ages between 120 and 130 days promoted the rate of 74% of regeneration. And S. polyrhiza 7498 showed 44% of regeneration of calli with ages between 45 and 55 days or between 85 and 95 days. From the results of antibiotic resistance experiments, where different concentrations of kanamycin, spectinomycin, streptomycin, geneticin, tobramycin and timentin were tested, it could be defined the lethal concentration for each of the three genotypes and which concentration allowed the growth of about 50% of plants, ideal concentration for use in plant selection in transformation experiments. The genetic transformation of the L. minor 8627 genotype resulted in the efficiency of 82,2% of GUS transient expression in calli and only 2,7% in fronds. Calli induction were obtained for the three genotypes, and also the plant regeneration from them. Calli of L. minor 8627 were transformed transiently.
