A biossíntese bacteriana de ácido indol-3-acético (AIA) exerce um papel fundamental na promoção do crescimento vegetal. Dentre as vias bacterianas de biossíntese de AIA, a via do indol-3-piruvato (IPyA) que tem como precursor o aminoácido L-triptofano (L-trp), regulada principalmente pelo gene ipdC, é a mais bem descrita em rizobactérias promotoras de crescimento em plantas (RPCP's). Apesar da importância, estudos moleculares relacionados às vias metabólicas da síntese bacteriana do AIA e dos genes envolvidos nesse processo são incipientes. A RPCP Bacillus sp. RZ2MS9, previamente isolada da rizosfera do guaranazeiro, vem apresentando satisfatória atividade promotora de crescimento vegetal. Desta promoção de crescimento vegetal destaca-se o desenvolvimento de raízes das plantas inoculadas, sugerindo-se a relevância do AIA produzido pela linhagem Bacillus sp. RZ2MS9 como mecanismo principal na interação bactéria-planta. Além disso, o sequenciamento de seu genoma apontou a presença de diversos genes relacionados à promoção e crescimento, com destaque para genes envolvidos na biossíntese de auxinas pela via IPyA. Assim, o presente estudo teve como objetivos: i) avaliar a influência da suplementação do aminoácido L-trp e do AIA na modulação da expressão dos genes da via IPyA: aaaT, ipdC e aldH em Bacillus sp. RZ2MS9, ii) avaliar os efeitos moleculares e fenotípicos do nocaute do gene ipdC na linhagem Bacilllus sp. RZ2MS9 ΔipdC, bem como seu efeito na interação com plantas de milho.

The bacterial biosynthesis of indole-3-acetic acid (IAA) plays a significant role in plant growth promotion. Among the bacterial pathways to IAA biosynthesis, the indole-3-pyruvate (IPyA), whose precursor is the amino acid tryptophan (Ltrp), regulated mainly by the ipdC gene, is the best described in plant growth promoting rhizobacteria (PGPR's). Despite its importance, metabolic pathways of bacterial synthesis of IAA and the genes involved in this process has not been studied well at a molecular level. The PGPR Bacillus sp. RZ2MS9, previously isolated from guaranazeiro rhizosphere, has been showing satisfactory plant growth promoting activity. The increment on root development by Bacillus sp. RZ2MS9 inoculation suggests the relevance of IAA during its plant interaction. Moreover, the sequencing of its genome indicated the presence of several genes related to growth promoting, with emphasis on genes involved in the biosynthesis of auxins through the IPyA pathway. Therefore, the present study has the following objectives: i) assess the influence of supplementation of the amino acid Ltrp and IAA on the expression of genes in the IPyA pathway: aaaT, ipdC and aldH in Bacillus sp. RZ2MS9; ii) evaluate the molecular and phenotypic effects of the knockout of the ipdC gene in the Bacilllus sp. RZ2MS9 ΔipdC as well as its interaction with maize plants.
