Introdução: o estudo de validação para verificar a repetitividade e reprodutibilidade com o uso de diferentes colorações ou sistema de marcação de bandas de proteínas da urina de cães, na eletroforese em gel de poliacrilamida, seria de grande importância pois alicerçariam informações fidedignas acerca da investigação da proteinúria de forma qualitativa e quantitativa, pela confiabilidade nos métodos e interpretação clínica adequada. Ainda, a possibilidade de comparação entre as colorações e sistema de marcação de bandas de proteínas poderá indicar o melhor procedimento laboratorial. Métodos: amostras de urina com diferentes concentrações de proteínas, denominadas como pools 1, 2 e 3, com valores da razão proteína creatinina urinária (RPC) de 0,04, 1,48 e 14,41, respectivamente, foram submetidas a avaliação do intra (repetição de 10 determinações no mesmo dia) e inter (repetição de 10 determinações no mesmo dia e repetidos consecutivamente em 3 dias seguidos) ensaios na eletroforese em gel de poliacrilamida no atinente aos pesos moleculares e porcentagem de densidade óptica das bandas. Foram empregadas as colorações pelo azul brilhante de Coomassie e pela Prata, como também pelo sistema Stain-Free®. Resultados: os coeficientes de variação para os pesos moleculares, no intra e inter ensaios, foram praticamente inferiores a 5% (variação de 0,41% a 6,55%) em todas as colorações e o Stain-Free®. Para a porcentagem de densidade óptica, o coeficiente de variação foi superior a 5% no intra (valor máximo de 28,95% na coloração do azul brilhante de Coomassie) e inter (valor máximo de 28,03%, também na coloração azul brilhante de Coomassie) ensaios e ambos na amostra de pool 3. Os coeficientes de variação foram maiores nas bandas de proteínas com maior porcentagem de densidade óptica, como também maiores na coloração Prata e coeficientes de variação menores para o sistema Stain-Free®. O sistema Stain-Free® foi o que apresentou com maior frequência diferença significante com as colorações azul brilhante de Coomassie e Prata. Conclusão: a repetitibilidade e a reprodutibilidade foram adequadas para a detecção das bandas dos pesos moleculares em todas as colorações e o Stain-Free®, não ocorrendo o mesmo para a porcentagem de densidade óptica, entretanto o sistema Stain-Free® foi o que apresentou os menores coeficientes de variação quando comparado as outras colorações.

Introduction: the validation study in order to verify the repeatability and reproducibility by the use of different stains or protein bands marker of urinary protein in dogs by sodium dodecyl sulfate (SDS) polyacrylamide gel electrophoresis is the great importance as those information could support reliable information by means of qualitative and quantitative investigation of proteinuria as well as to achieve adequate clinical interpretation due to the reliability of the methods. In addition, the possibility of the comparison among stains and protein bands marker could guide for better laboratorial procedures. Methods: urine samples of different concentrations of proteins called as pools 1, 2 and 3 showing urinary protein creatinine ratio (UPC) of 0.04, 1.48 and 14.41, respectively, were submitted to intra (repetition of 10 measurements on the same day) and inter (repetition of 10 measurements on the same day and that procedure repeated consecutively along 3 consecutive days) assays by the use of SDS polyacrylamide gel electrophoresis for the evaluation of molecular weights and percentage of densitometric analysis. Coomassie brilliant blue and silver stains were used as well as the Stain-Free® system. Results: the coefficients of variation for the molecular weights detected in the intra and inter assays were in its majority less than 5% (range of 0.41% to 6.55%) and were observed in all stains and Stain-Free® system. In relation to the percentage densitometric analysis of the protein bands, the coefficient of variation was greater than 5% in the intra assay (maximum value of 28.95% at the Coomassie blue stain) and inter assay (maximum value of 28.03% at Coomassie blue stain), and both observed in the pool 3 sample. The coefficients of variation were higher at protein bands with a higher percentage of densitometric analysis, and higher values were detected by the use silver stain and lower in the Stain-Free® system. The Stain-Free® system was one of the most that showed significant difference with the Coomassie brilliant blue and silver stains. Conclusion: repeatability and reproducibility were adequate for the detection of the protein molecular weight bands by the use of stains and Stain-Free® system, however not observed for the percentage of densitometric analysis of bands, and the Stain-Free® system showed the lowest coefficients of variation when compared to the others stains.
