Prototheca spp. é uma microalga unicelular que pode causar infecções oportunistas em humanos e animais. Este patógeno apresenta elevada resistência aos antimicrobianos convencionais e os atuais métodos de identificação de Prototheca spp. são demorados e com elevado custo. Diante disso, os objetivos do presente estudo foram (a): avaliar o uso da técnica de MALDI-TOF MS para identificação em nível de espécie de isolados de Prototheca spp. causadores de mastite bovina; (b) identificar a capacidade de produção de biofilme e (c) a sensibilidade in vitro de Prototheca spp. diante do polihexametileno biguanida (PHMB) e desinfetantes utilizados antes e após ordenha (hipoclorito de sódio, dicloroisocianurato de sódio, clorexidina e iodo-povidona). Um total de 87 isolados de Prototheca spp. foram submetidos a extração de proteínas ribossomais por beads de zircônia, e posteriormente identificados por MALDI-TOF MS pelo software Biotyper suplementado com espectros de referência. Para determinação do perfil de sensibilidade in vitro, os isolados de Prototheca spp. foram submetidos a avaliação de concentração inibitória mínima (CIM) e concentração algicída mínima (CAM) pelo método de microdiluição em caldo. A capacidade de formação de biofilme foi avaliada pelo método de incubação em microplacas de fundo chato e, posteriormente, os isolados foram classificados como fortes, moderados, fracos e não formadores de biofilme. A MALDI- TOF MS possibilitou a identificação de 73 (73/85) isolados como P. bovis e 2 (2/2) isolados como P. blaschkeae, sendo que, 12 (12/85) isolados de Prototheca foram identificados em nível de gênero. Do total dos isolados de P. bovis identificados por PCR, 63,5% (54/85) foram classificados como fortes, 28,2% (24/85) moderados, e 8,2% (7/85) fracos produtores de biofilme. Por outro lado, um isolado de P. blaschkeaefoi classificado como fraco e o outro como moderado produtor de biofilme. A PHMB (CIM90: ≥2 μ g/ml e CAM90: ≥4 μ g/ml) e o gluconato de clorexidina (CIM90 e CAM90: ≥2 μ g/ml), apresentaram os menores valores de CIM e CAM, seguido de dicloroisocianurato de sódio (CIM90 e CAM90: ≥1.400 μ g/ml), hipoclorito de sódio (CIM90 e CAM90: ≥2.800 μ g/ml), e iodo-polivinilpirrolidona (CIM90 e CAM90: ≥3.200 μ g/ml). Concluímos que o uso da MALDI-TOF MS permite a rápida identificação de isolados de Prototheca spp. em nível de espécie. Mais de 90% dos isolados de P. bovis apresentaram elevada capacidade de formação de biofilme. Dentre os desinfetantes, PHMB e o gluconato de clorexidina apresentaram os menores valores para inibição e inativação dos isolados de Prototheca spp. Sendo assim, estes antimicrobianos podem ser utilizados como alternativa antes e após a ordenha para o controle da disseminação de Prototheca spp. no rebanho.

Prototheca spp. is a unicellular microalgae that can cause opportunistic infections in humans and animals. This pathogen has high resistance to conventional antimicrobials and the current identification methods for Prototheca spp. are time-consuming and presently elevate costly. Due this, the objectives of the present study were (a): to evaluate the use of the MALDI-TOF MS for species-level identification of isolates of mastitis- causing Prototheca spp.; (b) evaluate the biofilm production capacity and (c)evaluate the in vitro sensibility of Prototheca spp. against polyhexamethylene biguanide (PHMB) and disinfectants (sodium hypochlorite, sodium dichloroisocyanurate, chlorhexidine gluconate and iodine-polivinilpirrolidona). A total of 87 isolates of Prototheca spp. were subjected to ribosomal protein extraction by zirconia beads, and identified by MALDI- TOF MS by Biotyper software supplemented with reference spectra. To determine the in vitro sensibility, isolates of Prototheca spp. were submitted to evaluation of Minimal Inhibitory Concentration (MIC) and Minimal Algicidal Concentration (MAC) by broth microdilution method. The biofilm production capacity was evaluated by the method of incubation in flat-bottomed microplates. Then, the isolates were classified as strong, moderate, weak and non-biofilm producers, by optical density values. The MALDI-TOF MS allowed the identification of 73 (73/85) isolates as P. bovis and 2 (2/2) isolates as P. blaschkeae, with 12 (12/85) isolates of Prototheca being identified at the genus level. Of the total of P. bovis isolates identified by PCR, 63.5% (54/85) were classified as strong, 28.2% (24/85) moderate, and 8.2% (7/85) weak producers of biofilm. On the other hand, one isolate of P. blaschkeae was classified as weak and the other as moderate biofilm producer. PHMB (MIC90: ≥ 2 µg/ml and MAC90: ≥ 4 µg/ml) and chlorhexidine gluconate (MIC90 and MAC90: ≥ 2 µ g/ml) had the lowest MIC and MAC values, followed by sodium dichloroisocyanurate (MIC90 and MAC90: ≥ 1400 µ g/ml), sodium hypochlorite (MIC90 and MAC90: ≥ 2,800 µ g/ml), and iodine-polivinilpirrolidona (MIC90 and MAC90: ≥3,200 µ g/ml). We conclude that the use of MALDI-TOF MS allows the rapid identification of Prototheca spp. at the species level. More than 90% of the P. bovis isolates presented high capacity for biofilm production. Among the disinfectants, PHMB and chlorhexidine gluconate presented the lowest values for inhibition and inactivation of Prototheca spp. Therefore, these antimicrobials can be used as an alternative to control the Prototheca spp. in the herd.
