A Bioengenharia pulmonar pode ser uma ferramenta terapêutica em potencial para a obtenção de pulmões para transplante em pacientes em uma fase muito avançada de doenças respiratórias. No entanto, não há dados disponíveis para decifrar o papel específico desempenhado pelos diferentes estímulos biofísicos que caracterizam a fisiologia respiratória na bioengenharia pulmonar quando uma matriz pulmonar decelularizada é cultivada com células- tronco. Diante disso, o objetivo principal deste estudo foi verificar se os estímulos físicos de estiramento estático, mimetizando a fisiologia respiratória, junto ao laser de baixa intensidade, podem otimizar a recelularização pulmonar com células tronco mesenquimais. Para isso, os pulmões passaram pelo processo de descelularização seguido da quantificação dos componentes da matriz extracelular (MEC), microscopia eletrônica de varredura, imunohistoquimica e quantificação de DNA, comprovando a eficiência da descelularização e a preservação de colágeno I, III e IV; fibronectina, laminina e elastina, além das estruturas vasculares e alveolares, comprovando também que os pulmões descelularizados respeitou o critério de - 50 nanogramas por micrômetro de tecido. Na sequência, o isolamento e cultura de células-tronco derivadas da polpa de dente revelou a expressão de CD105 (SH2) e CD73 (SH3/4) sendo negativos para marcadores de linhagem hematopoiéticos CD34, e o cultivo com células epiteliais Beas-2B e epitelial alveolar humana A-549 foram usadas juntamente as células-tronco na recelularização de 3 pulmões descelularizados, permitindo compreender melhor os mecanismos biofísicos básicos que participam na regeneração pulmonar utilizando um gerador de fluxo continuo com pressão positiva e um Laser duo de baixa intensidade. Por fim, nossos resultados demonstram que as terapias com laser e CPAP, modularam os níveis de citocina e previniram a contaminação.

Lung bioengineering can be a potential therapeutic tool for obtaining lungs for transplantation in patients at a very advanced stage of respiratory diseases. However, there is no data available to decipher the specific role played by the different biophysical stimuli that characterize respiratory physiology in pulmonary bioengineering when a decellularized pulmonary matrix is cultured with stem cells. Therefore, the main objective of this study was to verify whether the physical stimuli of static stretching, mimicking respiratory physiology, together with low-level laser, can optimize pulmonary recelularization with mesenchymal stem cells. For this, the lungs went through the decellularization process followed by the quantification of the components of the extracellular matrix (MEC), scanning electron microscopy, immunohistochemistry and DNA quantification, proving the efficiency of decellularization and the preservation of collagen I, III and IV; fibronectin, laminin and elastin, in addition to the vascular and alveolar structures, also proving that the decellularized lungs met the criterion of - 50 nanograms per micrometer of tissue. Following, the isolation and culture of stem cells derived from tooth pulp revealed the expression of CD105 (SH2) and CD73 (SH3 / 4), being negative for hematopoietic lineage markers CD34, and culturing with Beas-2B and epithelial cells A-549 human alveolar epithelial cells were used together in the recelularization of 3 decellularized lungs, allowing a better understanding of the basic biophysical mechanisms that participate in pulmonary regeneration using a continuous flow generator with positive pressure and a low intensity Duo Laser. Finally, our results demonstrate that therapies with laser and CPAP, modulated cytokine levels and prevent contamination.
