O permutador Na+/H+ está envolvido na regulação do pH e do volume celular. Em epitélios esse permutador é responsável pela reabsorção transepitelial de NaHCO3 e NaCl. A estrutura molecular desse permutador foi descrita pela primeira vez em 1989 e desde então foram descritas seis isoformas diferentes de permutador Na+/H+ em tecidos de mamíferos (NHE 1-6). Neste trabalho descrevemos a identificação da isoforma 1 deste permutador em tecido renal de coelho e a sequência do promotor do gene NHE-1 nesta mesma espécie. Além disso descrevemos a clonagem do permutador Na+/H+ existente em células de guelras do caranguejo Casmagnathus granulata. Referentes a estudos a modulação genética e funcional, descrevemos amodulação de NHE-3 em túbulos proximais de ratos por proteína quinase C e por PTH, em animais cronicamente depletados de PTH ou tratados com doses suprafisiológicas de PTH. Finalmente, descrevemos o estado de expressão genética e funcional do NHE-3 em rins submetidos a estímulo hipertrófico 4 e 24 horas após nefrectomia unilateral

Na+/H+ exchangers are important for intracellular pH and cell volume regulation. In epithelial cells, are responsible for NaHCO3 and NaCl reabsorption. The primary structure of a Na+/H+ exchanger was first described in 1989 and, since them, six isoforms of this protein have been described in mammalian cells (NHE- 1-6). In this paper, we describe the cloning and identification of the Na+/H+ exchanger type 1 in rabbit kidney and we present the sequence of the promoter region of the NHE-1 gene in this same species. It is also described the process of cloning of a Na+/H+ exhanger present in gills of the brazillian crab, Casmagnathus granulata. Relative to genetic and functional regulation of Na+/H+ exchangers, we describe the NHE-3 regulation in renal proximal tubules by protein kinase C and PTH, in rats cronicaly depleted of PTH, or treated with supraphysiological doses of PTH. Finally, we describe the expression and the functional activity of NHE-3 in remnant kidneys
