Camundongos da linhagem BALB/ c foram infectados com 5x106 leveduras de S. schenckii e acompanhados durante vinte e oito dias de infecção. Através da contagem do número de UFC nos baços fígados verificou-se que a carga fúngica nos órgãos dos animais diminui drasticamente na segunda semana de infecção. O teste ELISA revelou que nos órgãos destes animais a citocina IL-10 produzida em níveis mais elevados durante toda a infecção, assim modulando o controle da infecção. O padrão de proteínas secretadas pelas células leveduriformes de S. schenckii quando cultivadas em meio de cultura líquido e visualizado através de SDS-PAGE, apresentou bandas com pesos moleculares entre 97kDa e 20,1 kDa. Uma banda de 70 kDa foi reconhecida pelos soros dos animais infectados somente após a segunda semana de infecção e persistiu até o final desta. Os níveis de Ig total produzidos contra o antígeno solúvel e a proteína de 70 kDa aumentaram na segunda semana infecção e os títulos mantiveram-se durante a infecção. A isotipagem dos anticorpos revelou a predominância dos isotipos IgG1 e IgG3 para os dois antígenos testados. Notou-se que o aumento dos níveis de Ig contra o exoantígeno e a proteína imunorreativa de 70kDa, assim como o reconhecimento desta proteína, ocorreu inversamente proporcional à diminuição da carga fúngica nos órgão dos animais infectados. As proteínas secretadas pelas leveduras de S. schenckii, principalmente a proteína de 70 kDa, são capazes de ativar uma resposta humoral e conseqüente produção de altos níveis anticorpos e inibir a fagocitose do fungo pelos macrófagos. Desta maneira, estes antígenos modulam a resposta imune na esporotricose e contribuem para a patogenicidade do fungo.

BALB/c mice had been infected with 5xl06 yeasts of S. schenckii and followed during twenty and eight days of infection. Through the counting of the number of UFC in the livers and spleens it was verified that the fungaI load in the organs of the animais decreased drastically in the second week of infection. The ELISA test disclosed that in the organs of these animaIs the cytokine IL-10 was produced in leveIs more raised during alI the infection, thus modulating the control of the infection. The protein standard secreted for the yeasts of S. schenckii when cultivated in liquid medium and visualized through SDS-PAGE, it presented bands with molecular weights between 97 and 20,1 kDa. A band of 70 kDa was recognized for the sera of the infected animals after the second week of infection and persisted until the end of this. The leveIs of total Ig produced against the exoantigen and the protein of 70 kDa had increased in the second week of infection and the titers had been remained during the infection. The isotyping of the antibodies disclosed the predominance of the IgG1 and IgG3 for two tested antigens. One noticed that the increase of the leveIs of Ig against antigens and the recognition of the protein of 70 kDa occurred parallel with the reduction of the fungal load in the organs of the infected animals. The proteins secreted by the yeasts of S. schenckii, mainly the protein of 70 kDa, they are capable to activate a humoral response and consequent production high leveIs of antibodies and to inhibit phagocytosis of the fungi for the macrophages. In this way, these antigens modulate the immune response in sporotricosis and contribute for the pathogenic potential of fungi.
