Chromobacterium violaceum tem sido muito estudada a fim de explorar seu potencial biotecnológico, especialmente, a violaceína. Este trabalho teve por objetivo estabelecer estratégias para obtenção de mutantes. Linhagens mutantes foram obtidas por transposição com o mini-Tn5 e por recombinação homóloga (mutantes vioB e ubiB). O vioB em fase estacionária demonstrou ser mais resistente a UVC e aos sais metálicos FeCl2 e CdCl2. Com H2O2 não houve uma diferença significativa entre as taxas de sobrevivência exibidas pela linhagem vioB. Análise por microscopia demonstrou maior agregação das células do mutante vioB e uma maior formação de biofilme. O mutante ubiB apresentou crescimento deficiente na presença de 0.4 mg/mL de fenol.

Chromobacterium violaceum has been extensively studied for its biotechnological potential, especially what concerns violacein. In this work we have as goal to establish strategies for obtaining mutant strains. Mutant strains were obtained by transposition using mini-Tn5 and employing homologous recombination (mutant vioB and ubiB). vioB was shown to be more resistant in stationary phase to UVC and the metallic salts FeCl2 and CdCl2. With H2O2, there was no significant difference in survival rates exhibited by vioB. Microscopy analyses demonstrated a higher cell aggregation by the vioB mutant and a higher formation of biofilm. ubiB mutant presented growth defects in presence 0.4 mg/mL phenol.
