Alguns fungos na agricultura são classificados como fitopatógenos por causarem diversos danos a vários tipos de plantações, gerando doenças desde as raízes até os seus frutos. Uma das formas mais utilizadas para controlar tais patógenos é o emprego de agroquímicos como fungicidas. Entretanto, a utilização destes compostos químicos em excesso acarreta uma série de problemas como, por exemplo, contaminações desde o solo até os lençóis freáticos, além dos problemas que podem ocasionar à saúde do ser humano, direta e indiretamente. Estudos mostram que bactérias do gênero Bacillus produzem metabólitos secundários, os quais apresentam atividade antifúngica, entre outras, deste modo, apresentando-se como um potencial alternativa para substituição dos agroquímicos. O objetivo deste trabalho foi de identificar e caracterizar os metabólitos secundários secretados por um isolado da bactéria Bacillus velezensis, com ação antifúngica contra a Trichoderma reesei, Fusarium graminearum, F. oxysporum, F. verticillioides, Aspergillus flavus, Bothrytis sp. e Moniliophthora perniciosa. Realizou-se a identificação molecular da bactéria isolada com base na sequência da região 16S rDNA e fez-se uso do método de reconstrução filogenética neighbor-joining (NJ), que permitiu identificar a bactéria como Bacillus velezensis. Ensaios antagonísticos mostraram que a bactéria provoca inibição do crescimento dos fungos supracitados. Com intuito de determinar a estrutura do metabólito secundário responsável pela inibição do crescimento fúngico, o sobrenadante do cultivo da bactéria em caldo BD foi extraído com clorofórmio. Através da técnica de cromatografia em camada delgada preparativa (CCDP) foi possível obter uma fração de metabólitos que, após análises por UPLC-MS, revelaram-se como uma mistura de compostos. A análise desta mistura por ESI-CID/MS permitiu identificar três moléculas conhecidas de surfactina que apresentam cadeias de ácido graxo diferindo entre si no número de átomos de carbono, além da descoberta da estrutura de uma nova molécula ainda não descrita na literatura. O sobrenadante do meio de cultura foi precipitado com ácido, produzindo uma mistura de metabólitos que foram fracionados usando cromatografia de permeação em gel. Este procedimento forneceu quatro grupos de frações denominados FrSph 1, FrSph 2, FrSph 3 e FrSph 4, que foram analisados por UPLC-ESI-IT-MS e experimentos de ESI-MSn. Esses dados revelaram a presença concomitante de quatro moléculas de surfactina previamente relatadas com a adição de quatro metabólitos secundários da família da iturina na fração FrSph1. De outra forma, na fração FrSph 2 foram observados os mesmos compostos com predominância de três derivados de iturina e em menor teor duas surfactinas. A atividade antifúngica de todos os grupos de frações foram testadas por difusão em ágar com sete linhagens diferentes e o ensaio revelou a maior atividade observada para FrSph 1 e FrSph 2. A fração FrSph 1 mostrou atividade contra Trichoderma reesei, Fusarium graminearum, F. oxysporum, F. verticillioides, Bothrytis sp. e Moniliophthora perniciosa, mas não contra Aspergillus flavus, já a fração FrSph 2 mostrou atividade contra Trichoderma reesei, Fusarium graminearum, F. oxysporum, F. verticillioides, Aspergillus flavus e Bothrytis sp., mas não contra M. perniciosa. Esses resultados revelaram a importância do sinergismo entre os derivados de surfactina e iturina para a ação inibitória.

Some fungi in agriculture are classified as phytopathogens because they cause various damages to various types of plantations, generating diseases from the roots to their fruits. One of the most used ways to control such pathogens is the use of agrochemicals as fungicides. However, the use of these chemicals in excess entails a number of problems, such as contamination from soil to groundwater, as well as the problems that can directly and indirectly cause human health. Studies show that bacteria of the genus Bacillus produce secondary metabolites, which have antifungal activity, among others, thus presenting itself as a potential alternative for the replacement of agrochemicals. The objective of this work was to identify and characterize the secondary metabolites secreted by a Bacillus velezensis isolate, with antifungal action against Trichoderma reesei, Fusarium graminearum, F. oxysporum, F. verticillioides, Aspergillus flavus, Bothrytis sp. and Moniliophthora perniciosa. The molecular identification of the bacterium isolated on the basis of the sequence of the 16SrDNA region was carried out and the neighbor-joining (NJ) phylogenetic reconstruction method was used to identify the bacterium as Bacillus velezensis. Antagonistic assays have shown that the bacterium causes inhibition of growth of the aforementioned fungi. In order to determine the structure of the secondary metabolite responsible for the inhibition of fungal growth, the culture supernatant of the bacterium in BD broth was extracted with chloroform. Using the preparative thin-layer chromatography technique (PTLC) it was possible to obtain a fraction of metabolites which, after analysis by UPLC-MS, proved to be a mixture of compounds. Analysis of this mixture by ESI-CID/MS allowed us to identify three known surfactin molecules with fatty acid chains differing in the number of carbon atoms, in addition to new molecule not yet described in the literature. The supernatant of the culture medium was precipitated with acid, producing a mixture of metabolites whose were fractionated using gel permeation chromatography. This procedure furnished four fractions groups named as FrSph 1, FrSph 2, FrSph 3 and FrSph 4, which were analyzed by UPLC-ESI-IT-MS and ESI-MSn experiments. These data revealed the concomitant presence of four surfactin molecules previously related with addition of four iturin metabolites family in the FrSph1 fraction. The other side, in the fraction FrSph 2 were observed the same compounds with the predominance of three iturin derivatives and a lower content of two surfactins. The antifungal activity of all fractions groups by agar diffusion with seven different strains showed the greater activity observed for FrSph 1 and FrSph 2 fraction. The FrSph 1 fraction showed activity against Trichoderma reesei, Fusarium graminearum, F. oxysporum, F. verticillioides, Bothrytis sp. and Moniliophthora perniciosa, but not against Aspergillus flavus and the FrSph 2 fraction showed activity against Trichoderma reesei, Aspergillus flavus, Fusarium graminearum, F. oxysporum, F. verticillioides, and Bothrytis sp., but not against Moniliophthora perniciosa. These results revealed the importance of the synergism between surfactin and iturin derivatives to the inhibitory action.
