Curauá é uma planta cultivada na região da Amazônia e suas fibras lignocelulósicas contém um alto teor de celulose. Este estudo teve como objetivo contribuir com a diversificação de aplicações da fibra de curauá através da conversão da fração de polissacarídeos a açúcares fermentescíveis (sacarificação), por meio de hidrólise enzimática. Estes açúcares posteriormente podem ser convertidos no chamado etanol celulósico, através de fermentação. As fibras de curauá usadas neste estudo apresentaram como composição em torno de 76% de α-celulose, 16% de hemiceluloses e 8% de lignina Klason total. Fibras ricas em celulose, como as de curauá, têm ótimo potencial como material de partida para conversão a açúcares (neste caso glicose) fermentescíveis. Pré-tratamentos podem aumentar a eficiência desta conversão, através da modificação de propriedades das fibras como dimensões, morfologia da superfície, teor de lignina e/ou hemiceluloses, cristalinidade. Estudos anteriores corresponderam a uma exploração inicial da fibra de curauá quanto ao processo de sacarificação, submetendo-a a pré-tratamentos com solução aquosa alcalina (NaOH 20%) a temperatura ambiente, durante 2 h, o que aumentou o rendimento de glicose em relação a fibra não pré-tratada. O presente estudo visou explorar condições relativamente brandas para pré-tratamentos usando solução aquosa ácida de ácido oxálico (0,9M), selecionado por poder ser obtido a partir de fontes naturais, assim como testar condições diferentes das já usadas para pré-tratamento alcalino. Fibras previamente lavadas com água a temperatura ambiente e a 70 ºC (visando remover resíduos ou sais aderidos a fibra), e então submetidas a extração com mistura de etanol e cicloexano 1:1 (visando remover ceras, terpenos e ácidos graxos) foram submetidas a pré-tratamentos utilizando solução de ácido oxálico (AO) 0,9M na proporção de 1:30 de massa de fibra por volume de solução (g/mL), durante 1,5h a temperatura ambiente (AC0915AM), 1,5 h a 60 ºC (AC091560) e 3,0 h a 60 ºC (AC093060), e a pré-tratamento utilizando solução de hidróxido de sódio 20% durante 2 h (NaOH202060) e 5 h a 60 ºC (NaOH205060). Estas fibras pré-tratadas com AO e NaOH202060 foram posteriormente submetidas a hidrólise enzimática. As fibras pré-tratadas ou não foram caracterizadas quanto ao teor de holocelulose, de α-celulose, hemiceluloses, lignina total, microscopia eletrônica de varredura (MEV), cristalinidade (DRX), análise termogravimétrica (TGA, DTG), dimensões, e os resultados indicaram as seguintes mais relevantes modificações comparativamente às fibras não tratadas: aumento no teor de holocelulose de 92,1±0,4% para 93,1±0,7% (AC0915AM), redução no teor de α-celulose de 76,3±0,1% para 70,1±0,1% (AC091560), aumento no teor de hemiceluloses de 15,9±0,4% para 21,9±0,6% (AC091560), , e variação em cristalinidade de 61% (fibra de partida) para 75% (AC093060), indicando consumo de cadeias de celulose presentes nos domínios não cristalinos durante o pré-tratamento. O teor de lignina não apresentou variação significativa Os resultados também indicaram que tanto ácido quanto a base têm preferência em atacar as superfícies das fibras, comparativamente às extremidades, diminuindo a espessura das mesmas. A microscopia eletrônica de varredura (MEV) mostrou mudanças na superfície das fibras após pré-tratamento com AO e com NaOH nas diferentes condições, indicando que o maior tempo de pré-tratamento e maior temperatura resultaram em maiores modificações na morfologia das superfícies das fibras. A hidrólise enzimática foi realizada utilizando o complexo enzimático Accellerase 1500 (Genecor) que possui exoglucanases, endoglucanases, hemicelulases e β-glucanases e foi conduzia em tampão de citrato (50mL tampão/g fibra, pH 5), durante 48 h. Durante a hidrólise enzimática, alíquotas foram retiradas do meio em determinados intervalos de tempo, e as fibras não reagidas foram separadas por filtração do licor contendo os açúcares hidrolisados. As fibras não reagidas foram caracterizadas conforme as fibras de partida. As curvas DTG mostraram diminuição na intensidade do pico referente à decomposição térmica da celulose (em torno de 375 ºC) em diferentes tempos de reação da hidrólise enzimática devido a conversão da mesma em glicose, e mostraram redução do "ombro" referente a decomposição das hemiceluloses, devido a hidrólise das mesmas durante a reação. Os resultados referentes à avaliação das dimensões das fibras indicaram que a enzima atuou preferencialmente a partir da superfície das mesmas, comparativamente às extremidades, pois as variações na densidade das fibras foram superiores em relação a espessura comparativamente ao comprimento. Os resultados de índice de cristalinidade (DRX) indicaram que a enzima teve preferência em consumir preferencialmente as cadeias de celulose de domínios não cristalinos. As análises dos licores por HPLC foram realizadas tanto para as fibras pré-tratadas quanto para as fibras não pré-tratadas (esta somente após 48h). Após 48 h de reação, utilizando 0,5 mL.g-1 de enzima, os rendimentos foram de 58,6% (fibra não pré-tratada), 72,0% (AC0915AM), 74,8% (AC091560), 77,4% (AC093060) e 71,6% (NaOH202060). Para as fibras AC091560, utilizando 1,5 mL.g-1 de enzima o rendimento aumentou para 81,9%, mas uma análise de viabilidade econômica é necessária para avaliar se o gasto com enzimas não se contrapõe ao aumento no rendimento. As micrografias (MEV) mostraram que durante o pré-tratamento o ácido atacou a superfície das fibras de partida, diminuindo as espessuras das fibras mais espessas e a cristalinidade de 74% para 65%. O aumento no rendimento de glicose em relação a fibra de partida indica que esses fatores facilitaram o acesso das enzimas às cadeias de celulose. O tratamento alcalino resultou em rendimento maior (71,6 %) em relação a estudos anteriores. No entanto, este resultado foi aquém do esperado, indicando que o aumento na cristalinidade de 61% (fibras não pré-tratadas) para 79% dificultou o acesso das enzimas às cadeias de celulose. O presente estudo aprofundou investigações prévias, levando a um importante conjunto de resultados que podem embasar futuros estudos.

Curauá is a plant grown in the Amazon region and its lignocellulosic fibers contain a high content of cellulose. The aim of this study was to contribute to the diversification of curauá fiber applications by converting the polysaccharide fraction to fermentable sugars (saccharification) through enzymatic hydrolysis. Cellulosic ethanol is obtained from the fermentation of these sugars. The curauá fibers used in this study had around 76% of α-cellulose, 16% of hemicelluloses and 8% of total lignin Klason. Cellulose-rich fibers, such as curauá, have excellent potential as a starting material for converting to fermentable sugars (glucose). Pre-treatments may increase the efficiency of this conversion by modifying fiber properties such as size, surface morphology, lignin and/or hemicelluloses contents, and crystallinity. Previous studies corresponded to an initial exploration of the curauá fibers for the saccharification process, subjecting them to pre-treatments with aqueous alkaline solution (NaOH, 20% ), , room temperature,for 2 h, which increased the yield of glucose compared to untreated fiber. The present study aimed to explore relatively mild conditions for pre-treatments with acidic aqueous solution of oxalic acid (0,9M), which can be obtained from natural sources, as well as to test conditions different from those already used for alkaline pretreatment. Fibers previously washed with water at room temperature and at 70° C (to remove residues or salts adhered to fiber). Thereafter the fibers were extracted with a mixture of ethanol and cyclohexane 1:1 (to remove waxes, terpenes and fatty acids). After that the fibers were subjected to pretreatments using 0.9M oxalic acid solution (AO) at the ratio of 1:30 (g/mL) during 1.5h at room temperature (AC0915AM), 1.5 h at 60 ºC (AC091560) and 3.0 h at 60 ºC (AC093060). The fiber was also pretreated with sodium hydroxide 20% during 2.0 h at 60 ºC (NaOH 202060) and during 5.0 h at 60 ºC (NaOH 205060). The AO pretreated fibers were subsequently subjected to enzymatic hydrolysis, as well as NaOH 202060. The fibers were evaluated as to hollocelulose, α-cellulose, hemicelluloses and total lignin contents, crystallinity (DRX), thermogravimetric analysis (TGA, DTG), scanning electron microscopy (SEM). The results indicated the following most relevant modifications compared to untreated fiber: increase in holocellulose content from 92,1±0,4% to 93,1±0,7% (AC0915AM), reduction in α-cellulose content of 76,3±0.1% to 70,1±0,1% (AC091560), increase in hemicelluloses content from 15,9±0,4% to 21,9±0,6% (AC091560), and crystallinity variation from 61% to 75% (AC093060), indicating preferential consumption of the cellulose chains present in the non-crystalline domains of the fiber during pretreatment No significant change was observed for the lignin content. The results also indicated that both acid and base have a preference in attacking the surfaces of the fibers compared to the ends, decreasing their thickness. SEM showed changes in the surface of the fibers after pre-treatment with AO and NaOH under different conditions, indicating that the longer pretreatment time and higher temperature resulted in greater modifications in the fiber surface morphology. Enzymatic hydrolysis was performed using the enzyme complex Accellerase 1500 (Genecor) having exoglucanases, endoglucanases, hemicellulases and β-glucanases and was run in citrate buffer (50mL buffer/g fiber, pH 5) for 48 h. During the enzymatic hydrolysis, aliquots were withdrawn from the medium at certain time intervals, and the unreacted fibers were separated by filtration of the liquor containing the hydrolyzed sugars. The unreacted fibers were characterized according to the starting fibers. The DTG curves showed a decrease in the intensity of the peak relative to the thermal decomposition of the cellulose (around 375 ºC) in different reaction times of the enzymatic hydrolysis due to the conversion of the same into glucose, and showed reduction of the "shoulder" referring to the decomposition of hemicelluloses, due to their hydrolysis during the reaction. The results concerning the evaluation of the fiber dimensions indicated that the enzyme acted preferentially from the surface of the fibers, compared to the ends, since the variations in fiber density were higher in relation to the thickness compared to the length. The crystallinity index results indicated that the enzyme consumed preferentially cellulose chains of non-crystalline domains. Analysis of the liquors were performed by HPLC for both pretreated and non-pretreated fibers (this one only after 48h). After 48 h of reaction using 0.5 mL.g-1 enzyme, the yields were 58.6% (non-pretreated fiber), 72.0% (AC0915AM), 74.8% (AC091560), 77.4% (AC093060) and 71.6% (NaOH202060). For AC091560 fibers, using 1.5 mL.g-1 of enzyme the yield increased to 81.9%, but an economic viability analysis is required to assess whether the enzymes expense does not counteract the increase in yield. The micrographs (SEM) showed that during the pretreatment the acid attacked the surface of the starting fibers, decreasing the thicknesses of the thicker fibers, and the crystallinity from 74% to 65%. The increase in glucose yield relative to the starting fiber indicates that these factors facilitated the access of the enzymes to the cellulose chains. Alkaline treatment resulted in higher yield (71.6%) than in previous studies. However, this result was lower than expected, indicating that the increase in crystallinity from 61% (non-pretreated fibers) to 79% disfavored the access of the enzymes to the cellulose chains. The present study deepened previous investigations, leading to an important set of results that may support future studies.
