O Brasil é o maior consumidor de agrotóxicos do mundo e o desenvolvimento econômico no país está voltado principalmente para as atividades agrícolas, sendo o maior exportador de açúcar e o segundo maior exportador de soja no cenário mundial. Além dessas também se destacam as culturas de citrus, milho, algodão e tomate de maneira que a aplicação de agrotóxicos no combate aos seres considerados prejudiciais às plantações se mostra eficaz. No entanto, o uso indiscriminado dos agrotóxicos pode resultar em acumulação nos compartimentos ambientais, problemas de contaminação de águas, interferir na longevidade de organismos não-alvos, entre outros problemas. Assim, estudar a degradação de agrotóxicos se mostra de grande importância e relevância atualmente e este trabalho teve como objetivo estudar a biodegradação dos pesticidas Bifentrina e Fipronil por bactérias do gênero Bacillus isoladas de folhas da cultura de laranja. A Bifentrina é um pesticida classificado como piretróide e amplamente empregada em diferentes culturas. Uma triagem com as 8 linhagens de bactérias foi feita e por meio de análises por cromatografia líquida de alta eficiência (HPLC-UVis) verificou-se uma biodegradação da Bifentrina entre 79% e 88% pelas bactérias identificadas como B. pseudomycoides (2RF2C, 3RF2C, 4RF1C e 4RF2C) e as bactérias B. amyloliquefaciens (RFD1C), B. safensis/ B. pumilus (2RF1C), B. aryabhattai/ Bacillus sp. (2RF3C), B. megaterium (RF1C) apresentaram uma biodegradação da Bifentrina entre 25% e 38%. A bactéria B. pseudomycoides (4RF1C) apresentou uma degradação de 79% da Bifentrina e foi selecionada para um experimento em replicata para que os possíveis produtos de biodegradação fossem isolados e identificados. Por meio da técnica de cromatografia em coluna foi possível identificar o composto 4-hidroxibenzoato de metila, um possível metabólito produzido pela bactéria B. pseudomycoides (4RF1C). Das análises em cromatografia a gás acoplado à espectrometria de massas (CG-EM) foram identificados os compostos 4-di-terc-butilfenol, 1-nonadeceno, N-fenetilacetamida e 1,4-diisopropilbenzeno nas reações com a Bifentrina. Uma reação de hidrólise da Bifentrina com KOH foi realizada e os produtos da reação (o álcool BF-OH e o ácido BF-COOH) foram analisados por HPLC-Uvis verificar a presença desses compostos nas reações entre as bactérias e a Bifentrina. Em seguida, foi realizada uma reação utilizando o álcool BF-OH com a bactéria B. pseudomycoides (4RF1C) para observar possíveis produtos de biodegradação. Tal reação foi feita em 3 e 5 dias e a porcentagem de consumo do álcool BF-OH foi de 14% e 17%, respectivamente e não foi possível observar bandas de produtos de biodegradação para o álcool BF-OH. Um experimento de consórcio empregando as 8 linhagens de bactérias mostrou que a porcentagem de biodegradação do pesticida Bifentrina foi de 51%.  O Fipronil é um pirazol e apresenta aplicabilidade como herbicida, inseticida, fungicida e acaricida. Uma triagem com 8 linhagens de bactérias foi empregada para verificar a capacidade de degradação do pesticida Fipronil pelos microrganismos e, dentre as linhagens, as bactérias B. amyloliquefaciens (RFD1C) e B. aryabhattai/ Bacillus sp. (2RF3C) apresentaram a menor concentração de Fipronil depois de 5 dias de reação, com 93% e 91% de biodegradação ou biotransformação, ou seja, foram os microrganismos responsáveis pelo maior consumo do pesticida em meio líquido. Um experimento para avaliar a diminuição da concentração do pesticida Fipronil em 1, 3 e 5 dias também foi realizado, e um experimento de consórcio empregando as 8 linhagens de bactérias mostrou que a porcentagem de biodegradação do pesticida Fipronil foi de 81%. Por meio das análises de CG-EM foram identificados diversos compostos dentre os quais o terpeno esqualeno, o composto 1H-indol, a ciclo (Pro-Leu), N-fenetilacetamida, 5-hexil-di-hidrofurano-2-(3H)-ona, 1,3-diisopropilbenzeno e 1,4-diisopropilbenzeno. Análises no espectrômetro de massas de alta resolução acoplado ao cromatógrafo líquido de ultra-performance (UPLC-HRMS) foram feitas e observou-se possíveis metabólitos de biotransformação da Bifentrina e a Fipronil-sulfona proveniente da oxidação do Fipronil. Destaca-se também que foi identificado o terpeno esqualeno, possivelmente devido à associação entre a cultura de citrus e os microrganismos associados. Em todos os experimentos foram feitas reações controle na ausência do pesticida e mostraram boa recuperação quando quantificadas. Também foi feito um experimento para se avaliar a recuperação final tanto da Bifentrina como do Fipronil obtendo-se uma porcentagem de 96% de recuperação final para ambos os pesticidas. Por fim, este estudo demonstrou que as bactérias do gênero Bacillus isoladas das folhas da cultura da laranja foram importantes na degradação dos pesticidas Bifentrina e Fipronil utilizados em diferentes fases das culturas em que são aplicados.

Brazil is the largest consumer of pesticides in the world and the economic development in the country is mainly focused on agricultural activities, being the largest exporter of sugar and the second largest exporter of soybeans in the world scenario. In addition, citrus, corn, cotton and tomato crops stand out, so that the application of pesticides in the fight against beings considered harmful to crops is effective. However, indiscriminate use of pesticides can result in accumulation in environmental compartments, water contamination problems, to interfere with the longevity of non-target organisms, among other problems. Thus, studying the degradation of pesticides is of great importance and relevance today and this work aimed to study the biodegradation of pesticides Bifenthrin and Fipronil by bacteria of the genus Bacillus isolated from leaves of orange culture. Bifenthrin is a pyrethroid-classified pesticide widely used in different crops. Screening of the 8 bacterial strains was performed and by high performance liquid chromatography (HPLC-UVis) analysis, Bifenthrin biodegradation was found to be 79% to 88% by the bacteria identified as B. pseudomycoides (2RF2C, 3RF2C, 4RF1C and 4RF2C) and the bacteria B. amyloliquefaciens (RFD1C), B. safensis / B. pumilus (2RF1C), B. aryabhattai / Bacillus sp. (2RF3C), B. megaterium (RF1C) showed a Bifenthrin biodegradation between 25% and 38%. The bacterium B. pseudomycoides (4RF1C) showed a 79% degradation of Bifenthrin and was selected for a replicate experiment so that possible biodegradation products were isolated and identified. Using the column chromatography technique it was possible to identify methyl 4-hydroxybenzoate, a possible metabolite produced by the bacterium B. pseudomycoides (4RF1C). Gas chromatography-mass spectrometry (GC-MS) analyzes identified the compounds 4-di-tert-butylphenol, 1-nonadecene, N-phenethylacetamide and 1,4-diisopropylbenzene in reactions with Bifenthrin. A hydrolysis reaction of Bifenthrin by KOH was performed and the reaction products (alcohol BF-OH and acid BF-COOH) were analyzed by HPLC-Uvis to verify the presence of these compounds in the reactions between bacteria and Bifenthrin. Then, a reaction was performed using alcohol BF-OH with the bacterium B. pseudomycoides (4RF1C) to observe possible biodegradation products. This reaction was performed at 3 and 5 days and the percentage of alcohol BF-OH consumption was 14% and 17%, respectively, and it was not possible to observe bands of biodegradation products for alcohol BF-OH. A consortium experiment employing the 8 bacterial strains showed that the percentage of biodegradation of the pesticide Bifenthrin was 51%. Fipronil is a pyrazole and has applicability as herbicide, insecticide, fungicide and acaricide. A screening with 8 bacterial strains was used to verify the degradability of the pesticide Fipronil by the microorganisms and, among the strains, the bacteria B. amyloliquefaciens (RFD1C) and B. aryabhattai / Bacillus sp. (2RF3C) presented the lowest concentration of Fipronil after 5 days of reaction with 93% and 91% biodegradation or biotransformation, that is, they were the microorganisms responsible for the higher consumption of the pesticide in liquid medium. An experiment to evaluate the decay of the concentration of Fipronil pesticide at 1, 3 and 5 days was also performed, and a consortium experiment employing 8 bacterial strains showed that the percentage of biodegradation of the pesticide Fipronil was 81%. CG-MS analyzes identified several compounds including squalene terpene, 1H-indole, cyclo (Pro-Leu), N-phenethylacetamide, 5-hexyl dihydrofuran-2-(3H)-one, 1,3-diisopropylbenzene and 1,4-diisopropylbenzene. Analyzes on the high-resolution mass spectrometer coupled to the ultra-high performance liquid chromatograph (UPLC-HRMS) were performed and possible biotransformation metabolites of Bifentrin and Fipronil sulfone from Fipronil oxidation were observed. It is also noteworthy that squalene terpene was identified, possibly due to the association between citrus culture and associated microorganisms. In all experiments control reactions were performed in the absence of pesticide and showed good recovery when quantified. An experiment was also conducted to evaluate the final recovery of both Bifentrhrin and Fipronil to obtain a 96% final recovery for both pesticides. Finally, this study showed that bacteria of the genus Bacillus isolated from orange leaves were important in the degradation of pesticides Bifenthrin and Fipronil used in different stages of the crops in which they are applied.
