O ácido indol-3-acético (AIA) é uma auxina natural, metabólito do triptofano, e é produzido em células animal, vegetal e em alguns microrganismos podendo atuar como antioxidante. O objetivo do presente estudo foi o de avaliar o poder antioxidante do AIA, frente a um estresse oxidativo hepático induzido pelo hepatocarcinógeno dietilnitrosoamina (DEN). Foram utilizados camundongos BALB/c com dois meses. Em uma primeira etapa do estudo foram utilizados machos e fêmeas com o propósito de se avaliar somente o efeito do AIA administrado diariamente por 15 dias nas doses de 100, 200 e 500 mg/Kg de peso vivo. Em uma segunda etapa foram utilizados somente machos submetidos ao AIA (50, 250 e 500 mg/Kg de peso vivo) por 15 dias e no 16o dia foram expostos ao DEN por 4 h ou por 24 h. Foram avaliados os seguintes parâmetros: 1) evolução do peso e porcentagem relativa do fígado; 2) alterações metabólicas hepáticas avaliadas pelas atividades de alanina aminotransferase (ALT) e aspartato aminotransferase (AST) no soro; 3) perfil oxidativo hepático nas atividades das enzimas antioxidantes superóxido dismutase (SOD), catalase (CAT), glutationa peroxidase (GPx) e glutationa redutase (GR) e dos níveis de glutationa reduzida e glutationa oxidada e 4) fragmentação de DNA hepático. Os resultados foram analisados pela análise de variância (ANOVA) com significância de 0,05 usando o teste de Tukey.O AIA não demonstrou efeito tóxico nas doses utilizadas tanto para machos quanto para fêmeas em nenhum dos parâmetros analisados, pois esses parâmetros não apresentaram diferença significativa em relação aos controles (apenas tampão fosfato salina, pH 7,4). A exposição dos animais ao DEN por 4 h demonstrou diminuição na atividade da CAT e GR em 30% e 23%, respectivamente em relação ao controle. O DEN induziu fragmentação de DNA, somente após 24 horas, correspondentes a 210, 410 e 610 pares de bases (pb). A atividade da CAT e GR nos animais que receberam AIA e expostos ao DEN por 4 h não apresentaram alteração significativa em relação aos controles. Porém, a administração de AIA protegeu, apenas parcialmente o DNA dos efeitos defragmentação causados pela exposição ao DEN por 24 horas, pois se observou uma redução dessa fragmentação em 42%, 51% e 48% para os fragmentos de 210 pb, em 54%, 56% e 55% para os fragmentos de 410 pb e em 51%, 57% e 53% para os fragmentos de 610 pb nos animais que receberam AIA nas respectivas doses de 50, 250 e 500 mg/Kg de peso vivo, em relação aos controles. Concluiu-se que o AIA nas doses administradas, não alterou a evolução do peso, massa relativa do fígado, atividade de ALT e AST e perfil oxidativo hepático dos animais, sugerindo que esta auxina, não é tóxica; e que o AIA promoveu a proteção do fígado dos animais frente aos efeitos deletérios causados pelo DEN, baseado na atividade das enzimas CAT e GR e fragmentação de DNA, demonstrando um efeito antioxidante dessa auxina.

The indol-3-acetic acid (IAA) is a natural auxin, metabolite of the tryptophane, is produced in animal cells, vegetable and in some microorganisms being to act like antioxidant. The objective of the present study was to evaluate the antioxidant effect of the IAA, facing a liver oxidative stress induced by the hepatocarcinogenic diethylnitrosoamine (DEN). Were utilized mice BALB/c with 2 months. In a first phase of the study were utilized males and females with the purpose of to evaluate only the effect of the IAA administered daily by 15 days in the doses of 100, 200 and 500 mg/Kg of body weight. In a second phase were utilized only males submitted to IAA (50, 250 and 500 mg/Kg of body weight) daily during 15 days and in the 16ht day the animals were exposed to the DEN for 4 h or for 24 h. They were evaluated the following parameters: 1) evolution of the weight of the animals and the relative percentage of the liver; 2) liver metabolic alterations evaluated by the activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in the serum; 3) liver oxidative stress available by antioxidant enzyme activity as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione reductase (GR) and by levels of the oxidized glutathione and reduced glutathione and 4) fragmentation of liver DNA. The results were analyzed by the analysis of variance (ANOVA) with significance at 0.05 using Tukey test. The IAA did not showed toxic effect in the doses utilized for both males and females for all analyzed parameters, those parameters did not shown significant difference to compare with the controls (only phosphate buffer saline, pH 7.4). The exposition of the animals to DEN for 4 h showed a reduction in the CAT and GR activity as 30% and 23%, respectively, compared with to the control. The DEN to induced DNA fragmentation, only after 24 hours of exposition, corresponding to 210, 410 and 610 pair of bases (pb). The activity of the CAT and GR in the animal treated with IAA and exposed to DEN for 4 h did not show significant alteration to compare with the controls. However, the IAA administration was shown a partially protection of the DNA fragmentation from DEN exposition for 24 hours such as 42%, 51% and 48% for the fragments of 210 pb, 54%, 56% and 55% for the fragments of 410 pb and in 51%, 57% and 53% for the fragments of 610 pb by respectively IAA doses 50, 250 and 500 mg/Kg of body weight, compared with the controls. Concluded that the IAA in the doses administered, did not alter the evolution of the body weight, relative percentage of the liver, activity of ALT and AST and liver oxidative stress in animals, suggesting that this auxin is not toxic; and that the IAA promoted the protection of the liver of the animal facing the harmful effects caused by the DEN based in the activity of the CAT and GR activity and DNA fragmentation, showing an antioxidant effect of this auxin.
