A Própolis Verde Brasileira (PV) é um importante produto apícola, que apresenta importantes atividades biológicas, tornando-se valiosa no mercado internacional. O principal composto fenólico prenilado na PV é o artepelin C, juntamente com o seu ácido precursor, o ácido p-cumárico, ambos contribuem para os efeitos biológicos da PV. Levando isso em conta, avaliou-se o efeito da luz, temperatura e oxigênio do ar no teor destes compostos para se estabelecer as melhores condições de armazenamento e transporte para a PV bruta e para os compostos puros. Também foram avaliados o efeito do artepelin C, do ácido p-cumárico e seus produtos de degradação majoritários contra sete linhagens de bactérias orais, linhagens de células cancerígenas (AGP-01 e HeLa) e L. amazonensis (promastigotas e amastigotas). Para tanto, o artepelin C e o ácido p-cumárico foram submetidos à degradação por cinco dias sob luz solar e temperatura de 50 ºC, fornecendo três isômeros principais do artepillin C e um do ácido p-cumárico. Em seguida, foi desenvolvido e validado um método em Cromatografia Líquida de Alta Eficiência de Fase Reversa (CLAE-FR) para quantificação destes compostos na PV bruta e em seus extratos. Nos estudos de estabilidade utilizou-se o Design Experimental Fatorial Completo e o Design Composto Central Rotacionado para se estabelecerem as condições desejáveis de armazenamento da própolis bruta e seus metabólitos. Os ensaios biológicos foram realizados in vitro. O artepelin C, puro e na PV deve ser mantido protegido da luz e armazenado a temperaturas inferiores a -2,5 ºC para se evitar degradação. O ácido p-cumárico pode ser armazenado à temperatura ambiente. Com relação aos efeitos biológicos, consideraram-se promissores os compostos com valores de IC50 abaixo de 10 µM, sendo que o ART4 e o isômero Z do artepelin C apresentaram efeito significante contra a linhagem AGP-01 de câncer gástrico e contra Leishmania amazonensis, especialmente contra as formas amastigotas. O ácido p-cumárico também apresentou atividade promissora contra amastigotas de L. amazonensis, enquanto o AC-1, seu isômero Z, foi eficaz contra as formas promastigotas. Estes compostos não foram citotóxicos para CHO-k1, uma linha celular normal, indicando que não são tóxicos para as células normais. Portanto, as melhores condições de armazenamento e transporte para se manter o teor de ambos os compostos na PV são proteção da luz e armazenamento a baixas temperaturas. Além disso, esses compostos são candidatos promissores para o desenvolvimento de novos fármacos contra leishmaniose e câncer gástrico.

Brazilian Green Propolis (BGP) is an important bee product because it displays important biological activities. Therefore, it's valuable in the international market. The major prenylated phenolic compound in BPG is (E)-artepillin C, along with its precursor (E)-p-coumaric acid, and they contribute to the biological effects of BGP. Taking that into account, it was evaluated the effect of light, temperature and air oxygen in their content to establish the best storage and transport conditions for both crude BGP and its isolated compounds. It was also evaluated the effect of artepillin C, p-coumaric acid, and their major degradation products against seven strains of oral bacteria, cancer cell lines (AGP-01 and He-La) and L. amazonensis (promastigotes and amastigotes). For that, artepillin C and p-coumaric acid were initially submitted to degradation for five days under sunlight and high temperature (50 ºC), furnishing three major artepillin C isomers and one p-coumaric acid isomer. Then, it was developed and validated a Reverse Phase High Performance Liquid Chromatography (RP-HPLC) method for quantifying these compounds in crude BGP and in its extracts. In the stability studies, it was used a Full Factorial and Central Composite Design to establish the desirable storage conditions. The biological assays were performed in vitro. Crude BGP and artepillin C should be kept protected from light and should be storage below -2.5 ºC to prevent degradation. p-Coumaric acid can be stored at room temperature. Regarding the biological effects, it was considered the compounds with IC50 values below 10 µM. ART4, the Z isomer of artepillin C, displayed significant effect against gastric cancer cell line AGP-01 and against Leishmania amazonensis, especially the amastigote form of the parasite. P-coumaric acid also showed a promising activity against L. amazonensis amastigotes, while AC-1, its Z-isomer, was effective against the promastigote forms. These compounds were not cytotoxic to CHO-k1, a normal cell line, indicating that they are not toxic to normal cells.Therefore, the best storage and transport conditions to keep the content of both compounds in BGP are protection from light at low temperatures. Also, these compounds are promising candidates for development of novel drugs and should be further evaluated.
