A pele atua como barreira entre o meio externo e o organismo. Mesmo possuindo um grande número de mecanismos de defesa antioxidante, certas situações como a exposição prolongada à radiação ultravioleta (RUV), são capazes de romper o equilíbrio pró-oxidante/antioxidante no organismo, provocando um grande aumento na concentração de espécies reativas de oxigênio (EROs). A administração tópica de antioxidantes é um eficiente modo para enriquecer o sistema protetor cutâneo endógeno, e assim uma estratégia de sucesso para reduzir os danos oxidativos causados à pele pela RUV. O objetivo do presente trabalho foi avaliar os extratos vegetais de Eugenia biflora, Eugenia protenta, Protium heptaphyllum, e Miconia minutiflora quanto as suas propriedades antioxidantes, bem como sua citotoxicidade, e verificar a eficácia do extrato de Protium heptaphyllum como agente fotoquimioprotetor in vivo. A atividade antioxidante dos extratos foi avaliada por diferentes métodos in vitro que simulam a formação de EROs e os valores foram expressos como a concentração de extrato necessária para promover 50% da ação antioxidante (IC50). Todos os extratos estudados tiveram valores de IC50 baixo, o que representa um grande potencial antioxidante. Além disso, o extrato de Protium heptaphyllum apresentou baixa citotoxicidade em concentrações antioxidantes e quando irradiado parece ser menos citotóxico. Assim, a eficácia fotoquimioprotetora deste extrato incorporado a uma formulação tópica foi avaliada por meio de estudos in vivo em camundongos sem pelos expostos à RUV. O extrato incorporado em uma formulação gel, capaz de disponibilizar seus ativos antioxidantes na pele dos animais, mostrou ser capaz de recuperar os níveis de GSH e a atividade da SOD reduzidos pela RUV. Por outro lado, não houve inibição da atividade das mieloperoxidases. Estes resultados indicam que o extrato apresenta potencial como agente fotoquimioprotetor

The human skin acts as barrier between the environment and the organism. It presents a large number of antioxidant defense mechanisms, however, in some situations, such as in prolonged exposure under ultraviolet radiation (UVR), it is possible to break the pro-oxidant/antioxidant equilibrium in organism, causing an increase of reactive oxygen species (ROS). Topic administration of antioxidants is an efficient way to enhance cutaneous defense system, and a strategy to reduce oxidative damages caused by UVR in the skin. The purpose of this study was to evaluate the antioxidant properties and the citotoxicity of Eugenia biflora, Eugenia protenta, Protium heptaphyllum and Miconia minutiflora plant extracts, and to test the in vivo photochemoprotective efficiency of Protium heptaphyllum plant extract. The antioxidant activity of plant extracts was evaluated by several in vitro methods that simulated ROS generation and the results were expressed such as the extract concentration necessary to promote 50% of the antioxidant action (IC50). All the studied plant extracts presented low IC50 values, showing a great antioxidant potential. Moreover, Protium heptaphyllum extract presented reduced citotoxicity in antioxidant concentrations and the exposure under UVR seems to decrease its citotoxicity. So, the photochemoprotective effectiveness of this extract into a topical formulation was evaluated by in vivo tests using hairless mice exposed under UVR. Gel formulation with Protium heptaphyllum extract proved to be able to provide antioxidant compounds to skin and to recover GSH levels and SOD activity reduced by UVR. On the other hand, there was no inhibition of myeloperoxidases activity. These results indicate that this extract presents potential as photochemoprotective agent.
