Nesta Dissertação de Mestrado, buscamos estudar o mecanismo da reação de fotólise por radiação ultravioleta (UV) do ácido 2-hidroxi-3,5-dinitrobenzoico (DNS) e do ácido 5-amino-2-hidroxi-3-nitrobenzoico (7). Foram usados como compostos modelos: o ácido 2- hidroxibenzoico (1), ácido 2-hidroxi-5-nitrobenzoico (2), ácido 2-hidroxi-3-nitrobenzoico (3), ácido 5-amino-2-hidroxibenzoico (4), ácido 5-nitroftálico (5-NFt), orto-nitrofenol (o-NF) e para-nitrofenol (p-NF). Inicialmente, preparamos o ácido 5-amino-2-hidroxi-3-nitrobenzoico (7) por uma rota sintética de cinco etapas. Depois, soluções aquosas 1,0 mmol L-1 do DNS, 7 e dos compostos modelos foram submetidas a fotólise por UV, durante 240 min. Periodicamente alíquotas foram recolhidas e analisadas por cromatografia líquida de alta eficiência acoplada à espectrometria de massas (CLAE-EM) com ionização em modo negativo (Electrospray Ionization - ESI(-)). Os resultados indicam a formação de produtos de: (a) hidroxilação mediada por solvente, (b) descarboxilação-hidroxilação, (c) descarboxilação e (d) dímeros, dependentes do tipo de substituinte e de sua posição no anel aromático. O possível mecanismo para gerar estes intermediários envolve, no início, um radical fenoxila produzido pela clivagem fotoquímica da ligação O-H da hidroxila fenólica presente em DNS, 7 e nos demais compostos estudados. Determinamos a entalpia da dissociação da ligação O-H da hidroxila fenólica por meio de reações isodésmicas calculadas por Teoria do Funcional de Densidade (DFT) com conjunto de base B3LYP/6-31+G(d,p). A clivagem fotoquímica da ligação hidroxila é fortemente influenciada pelo caráter doador ou captor de densidade eletrônica que o substituinte possui e por sua posição no anel aromático e gera o radical fenoxila. A análise de densidade de spin de Mulliken (ρ5) por DFT/B3LYP/6-31+G (d,p) confirma que o elétron desemparelhado no átomo de oxigênio do radical fenoxila deslocaliza-se pelos átomos de carbono nas posições 1, 3 e 5 do anel aromático e em átomos de nitrogênio de aminas. Isto justifica a formação de produtos intermediários identificados ao longo da fotólise por UV do DNS e demais compostos. A influência do radical fenoxila nas reações de fotólise por UV foi comprovada por experimentos de fotólise com 5-NFt, onde a hidroxila fenólica foi substituída por -COOH. Neste caso não houve a formação de produtos de hidroxilação mediada por solvente. O grupo - COOH é imprescindível nos compostos DNS, 7 e em seus análogos, pois enfraquece as ligações C-NO2 facilitando que processos de hidroxilação mediada por solvente ocorram. Isto é comprovado quando comparamos os resultados da fotólise por UV de 2 e 3 com o p-NF e oNF, respectivamente.

In this work, we investigated the mechanism of ultraviolet (UV) photolysis reaction of 2- hydroxy-3,5-dinitrobenzoic acid (DNS) and 5-amino-2-hydroxy-3-nitrobenzoic acid (7). We also used as model compounds: 2-hydroxybenzoic acid (1), 2-hydroxy-5-nitrobenzoic acid (2), 2-hydroxy-3-nitrobenzoic acid (3), 5-amino-2-hydroxybenzoic acid (4), 5-nitrophthalic acid (5- NFt), ortho-nitrophenol (o-NF) and para-nitrophenol (p-NF). First, we prepared 5-amino-2- hydroxy-3-nitrobenzoic acid (7) by a five-step synthetic route. Later, aqueous solutions 1.0 mmol L-1 of DNS, 7 and the model compounds were subjected to UV photolysis for 240 min. Periodically, aliquots were collected and analyzed by High Performance Liquid Chromatography coupled to Mass Spectrometry (HPLC-MS) with Electrospray Ionization in negative mode (ESI(-)). The results show the formation of products of: (a) solvent mediated hydroxylation, (b) decarboxylation-hydroxylation, (c) decarboxylation and (d) dimers, dependent on the type of substituent and its position in the aromatic ring. The possible mechanism for generating these intermediaries involves, at beginning, a phenoxyl radical produced by the photochemical cleavage of the phenolic O-H bond present in the DNS, 7 and in other studied compounds. We calculated the enthalpy of dissociation of the phenolic O-H bond through isodesmic reactions applying the Density Functional Theory (DFT) with B3LYP/base set 6-31+G(d,p). The electron donor or withdrawing characters of the substituents and their location in the aromatic ring strongly influence the hydroxyl bond photochemical cleavage for producing the phenoxyl radical. Mulliken spin density analysis (ρ 5) by DFT/B3LYP/6-31+G(d,p) validates that the unpaired electron of the phenoxyl radical´s oxygen relocates to carbon atoms at positions 1, 3 and 5 of the aromatic ring and to nitrogen atoms of amines. This justifies the formation of intermediary products identified during the UV photolysis of DNS and analogous compounds. The role of the phenoxyl radical on UV photolysis reactions was proven by photolysis experiments with 5-NFt, where the phenolic hydroxyl was replaced by -COOH. In this case, there was no formation of hydroxylation products mediated by solvent. The presence of the -COOH group in DNS, 7 and others models compounds is crucial, since it weakens the C-NO2 bonds, favoring the formation of solventmediated hydroxylation products. This is supported by the results from the UV photolysis of 2 and 3 compared to the p-NF and o-NF, respectively.
