Abstract in Portuguese
A utilização de biopolímeros na dentina desmineralizada tem sido analisada com o intuito de melhorar as propriedades mecânicas e estruturais da dentina previamente ao procedimento restaurador. Este estudo in vitro avaliou o efeito da reumificação da dentina com quitosana em diferentes concentrações sobre a resistência adesiva, composição química da dentina e morfologia da interface dentina/resina. Os fatores em estudo foram substrato dentinário em dois níveis: hígido e desmineralizado; e reumidificação da dentina em quatro níveis: quitosana a 2,5%, quitosana a 5,0%, ácido acético a 1,0% (placebo) e ausência de reumidificação (controle). Oitenta dentes bovinos foram seccionados no terço médio da coroa para obtenção dos espécimes de dentina (7,0 X 7,0 mm). Os espécimes foram planificados e polidos, e então foram divididos aleatoriamente em 8 grupos (n=10). Metade dos espécimes foi submetida à indução de cárie artificial pelo método de ciclagem de pH (8h em solução desmineralizante com pH 4.8 e 16 horas em solução remineralizante com pH 7.0) e então, foram subdivididos de acordo com a reumidificação da dentina. Os espécimes foram restaurados e o conjunto dente/restauração foi seccionado a fim de ser obter 6 palitos com área transversal de 1,0±0,2 mm2, que foram submetidos a análise quantitativa (MPa) da resistência de união a microtração. Três espécimes de cada grupo foram desidratados em grau ascendente de etanol e submetidos à análise dos níveis de cálcio e fósforo da superfície dentinária por meio de espectroscopia eletrônica de análise química (EDS) e análise qualitativa da interface adesiva por meio de microscopia eletrônica de varredura (MEV). Os resultados obtidos da análise de microtração foram submetidos a análise de variância (ANOVA) e apresentaram diferença estatisticamente significante para o tipo de substrato dentinário (p=0,00001) e para reumidificação da dentina (p=0,0105). O teste de Tukey encontrou que para o tipo de substrato, a dentina hígida apresentou maior resistência adesiva e que a dentina reumidecida com quitosana a 2,5% apresentou maior resistência adesiva quando comparada a quitosana a 5,0%, ácido acético a 1,0% ou ausência de reumidificação. O teste de Kruskal Wallis encontrou diferença significante para os níveis de cálcio (p=0,036) e fóforo (p=0,019) encontrados na análise de EDS. O teste de Mann-Whitney mostrou que em substrato desmineralizado a reumidificação com ácido acético a 1,0% e quitosana a 2,5% apresentou maiores concentrações de ambos os minerais. As imagens de microscopia eletrônica de varredura mostram partículas de quitosana no interior dos túbulos dentinários, sendo que a reumidificação da dentina desmineralizada com quitosana a 5,0% apresentou maior quantidade de partículas. A reumidificação da dentina com quitosana não influenciou a morfologia da interface dentina/resina, no entanto, a reumidificação da dentina desmineralizada com quitosana a 2,5% gerou alterações na composição química da dentina e proporcionou maior força de adesão.

The use of biopolymers in demineralized dentin has been investigated with the aim of improving the mechanical and structural properties of dentin prior to restorative procedure. The purpose of this study was to evaluate the rewetting of dentin with chitosan at different concentrations, on bond strength, chemical composition and morphology of dentin/resin interface. The factors under evaluation were: dentin substrate at two levels: sound and demineralized; and dentin rewetting at four levels - 2.5% chitosan, 5.0% chitosan, 1.0% acetic acid (placebo) and no rewetting (control). Eighty specimens of bovine dentin (7x7 mm) were randomly assigned into 8 groups (n=10). Half of dentin slabs were submitted to cariogenic challenge (8h in demineralizing solution and 16h in remineralizing solution) and then, were subdivided according to rewetting of dentin. The specimens were restored and each tooth was sectioned in order to obtain 6 sticks with cross-sectional area of 1.0±0.2 mm2, which were submitted to microtensile bond strength test (µTBS). Three specimens of each group were dehydrated in ethanol and submitted to analysis of calcium and phosphorus levels of dentin surface by electron spectroscopy of chemical analysis (EDS) and qualitative analysis of interface under scanning electron microscopy (SEM). For data obtained on microtensile bond strength test, ANOVA showed significant difference between dentin substrates (p=0.00001) and dentin rewetting (p=0.0105). Tukey test showed that for dentin substrate, sound dentin had greater bond strength than demineralized dentin and the rewetting with 2.5% chitosan provided higher bond strength than 5.0% chitosan, 1.0% acetic acid or no rewetting. Kruskal Wallis test found significant difference for calcium (p=0.036) and phosphorus (p=0.019) levels found in EDS analysis. Mann-Whitney test showed that in demineralized dentin, the rewetting with 1.0% acetic acid and 2.5% chitosan had higher concentration of both minerals. SEM analysis showed particles of chitosan within the dentinal tubules, specially when 5.0% chitosan were used. Dentin rewetting with chitosan did not changed the morphology of dentin/resin interface, however, the rewetting of demineralized dentin with 2.5% chitosan modified the chemical composition and provided better substrate for adhesive procedures, resulting in higher bond strength.
