O acúmulo de biofilme sobre a superfície dental pode gerar diversas complicações para a cavidade bucal, além de acometer a saúde geral do indivíduo. Assim, o objetivo deste trabalho foi avaliar in vivo a capacidade do emprego da irradiação com laser de Diodo no biofilme bacteriano dental maduro na inativação dos micro-organismos presentes. Foram selecionados 12 voluntários com acúmulo de biofilme sobre a superfície vestibular dos dentes. O presente estudo foi fatorial, com período de realização da irradiação em 2 níveis (controle/sem irradiação e após a irradiação) e o tipo de tratamento em 2 níveis (irradiação de laser de Diodo com 660 e 970 nm). Foram testados 4 sítios de cada voluntário, divididos aleatoriamente em 2 grupos/4 subgrupos experimentais. Inicialmente, foram coletadas 2 amostras iniciais do biofilme. Cada amostra foi inserida em um tubo Eppendorf correspondente contendo o fluido de transporte reduzido (RTF). Em seguida, previamente à irradiação, foi aplicada uma dose do fotossensibilizador azul de metileno sobre toda a superfície vestibular com um tempo de pré-irradiação de 5 minutos. A irradiação foi realizada sobre a superfície vestibular na forma de varredura. As amostras irradiadas foram coletas e dispersadas em um tubo Eppendorf correspondente. As amostras foram diluídas em RTF e semeadas em meio de cultura MM10 Sacarose Ágar em duplicata. Após 7 dias em anaerobiose à 37°C, foram contadas as unidades formadoras de colônias (UFC) de cada amostra sobre a superfície da placa. Foi realizada a comparação da quantidade de UFC/mg antes e após os tratamentos. Os dados foram analisados pelos testes de Wilcoxon e Mann Whitney com =0,05. Pode-se observar que ambos os tratamentos realizados foram capazes de diminuir estatisticamente (p<0,05) o número de micro-organismos presentes no biofilme bacteriano maduro, não havendo diferença significante entre eles. Conclui-se que os dois comprimentos de onda do laser de diodo apresentaram comportamentos similares, promovendo uma ação bactericida sobre o biofilme dental maduro

Biofilm is a highly structured community capable of causes many bacterial diseases that affects health and quality of life, causing several clinical conditions, such as dental caries. Therefore, the aim of this study was to evaluate in vivo the bactericidal ability of antimicrobial photodynamic therapy (aPDT) against a complex biofilm. It was selected twelve volunteers with biofilm accumulation over enamel surface. The presente study was factorial, the irradiation time was diveded in 2 levels (control/without irradiation and after irradiation) and the treatment in 2 levels too (irradiation with a Diode laser with 660 nm and 970 nm). Each volunteer had 4 samples tested, divided randomly into 2 groups/4 subgroups. Before treatments, 2 sample of complex biofilm were collected and stored in reduced transport fluid (RTF) to maintain the viability of the microorganisms. Thereafter, another two corresponding areas with biofilm were irradiated with Diode laser, according to their experimental groups. Which sample was collected and stored in RTF. The biofilm samples from all the experimental groups were diluted in the same RTF as they were stored. All of the samples diluted were cultured in culture media MM10 Sucrose Agar and incubated in anaerobic atmosphere at 37°C. After 7 days, the samples were analysed and the total of colony-forming units (CFU) was determined. A comparison of the CFU/mg of samples before and after treatments was made. Data were analysed using Wilcoxon and Man Whitney test (=0.05). Both treatments were able to statistically reduce (p<0.05) the number of microrganisms presentes at a complex biofilme, so there were no significant differences between them. The conclusion is that both wavelength of diode laser behaved in similar ways, acting as a bactericidal agent on a complex oral biofilm
