Introdução: As células tronco podem contribuir na patogênese da endometriose. Os objetivos deste estudo foram avaliar e comparar a expressão de genes relacionados à atividade das células tronco no endométrio tópico (Et), peritônio normal (PN) e nos implantes de endometriose peritoneal superficial e profunda (EPS/EPP) de mulheres inférteis com endometriose. Métodos: Vinte e quatro amostras de Et, PN, EPS e EPP foram obtidas durante laparoscopia de seis pacientes. A expressão de 84 genes relacionados à atividade de células tronco foi avaliada pela técnica de transcrição reversa e reação em cadeia de polimerase (RT-PCR). Resultados: Todos os genes foram expressos no Et, PN, EPS e EPP. Não houve diferença entre o PN, EPS e EPP. Não houve diferença quando as lesões de EPS e EPP foram comparadas entre si. Comparados ao Et, 24, 49 e 45 genes foram diferencialmente expressos no PN, EPS e EPP, respectivamente. Destes genes diferencialmente expressos 23 eram comuns. A análise funcional dos 23 genes evidenciou 5 genes comumente superexpressos (genes reguladores do ciclo celular; comunicação celular e marcador de células embrionárias) e 18 subexpressos (fatores de crescimento; marcadores de células neurais; marcador de auto-renovação; marcador de células embrionárias; divisão celular simétrica e assimétrica; marcadores de células hematopoiéticas; comunicação celular, via de sinalização Notch; via de sinalização Wnt; marcador de células mesenquimais; marcador metabólico e moduladores do cromossomo e da cromatina). Conclusões: Os genes relacionados à atividade de células tronco estavam expressos no endométrio tópico, nas lesões de endometriose peritoneal superficial e profunda e no peritônio normal. A expressão gênica foi mais semelhante no peritônio normal e nas lesões de endometriose peritoneal superficial e profunda, comparadas ao endométrio tópico. Não se observaram diferenças significantes na expressão gênica entre as lesões de endometriose peritoneal superficial e profunda

Introduction: Stem/progenitor cells may contribute to the pathogenesis of endometriosis. The objective of this study was to identify and compare expression of genes regulating SPCs function in eutopic endometrium (EuE), normal peritoneum (NP) and peritonea superficial and deep infiltrative endometriosis (SE/DIE) of women with pelvic endometriosis. Methods: Twenty-four biopsies from EuE, NP, SE and DIE were obtained during laparoscopy from 6 patients. The expression of 84 stem cell-related genes was performed using a RT-PCR-based analysis. Results: All genes analyzed were expressed in the EuE, NP, SE and DIE. No difference was shown between SE, DIE and NP. There was no difference when the SE and DIE were compared to each other. Compared to EuE, 24, 49 and 49 genes were differentially expressed in the NP, SE, and DIE, respectively. Of these differentially expressed genes 23 were the same. Funcional analisis demonstrated that 5 genes were commonly overexpressed (cell cycle regulators; cell communication and embryonic cell marker) and 18 underexpressed (growth regulators; neural cell markers; self-renewal marker; embryonic cell marker; symmetric and asymmetric stem cell division; hematopoietic cell markers; cell communication; Wnt pathway; Notch pathway; mesenchymal cell marker; metabolic marker; chromosome and chromatin modulators). Conclusions: Stem cell related genes were expressed in the human endometrium, peritoneal superficial and deep endometriosis lesions and in the normal peritoneum. There was no difference when superficial and deep endometriosis were compared to each other.
