A dependência com a temperatura da intensidade de luz espalhada por dispersões aquosas do lipídio aniônico DMPG (dimiristoil fosfatidilglicerol) foi estudada para diferentes forças iônicas. A transição principal, gel - líquido cristal, pode ser bem monitorada por um decréscimo abrupto na turbidez. Como esperado, a temperatura da transição de fase principal (Tm) aumenta com o aumento da força iônica. Para baixa força iônica, uma segunda transição com a temperatura para o DMPG, chamada de pós-transição, pode ser monitorada tanto por um aumento no espalhamento de luz quanto por um decréscimo na condutividade. Análises de Zimm plot indicam que abaixo de Tm os lipossomos tendem a se agregar, resultando em um segundo coeficiente de virial, A2, negativo, e em uma grande massa molecular da partícula. Na transição de fase, juntamente com uma diminuição na turbidez, ocorre um aumento na condutividade da amostra, A2 torna-se positivo e a massa molecular da partícula diminui, indicando um estado não agregado. Além disso, na pós-transição (Tp) A2 torna-se muito pequeno, e a massa molecular aumenta novamente. Ambas as transições são reversíveis. Opostamente à transição principal, a pós-transição não pôde ser detectada por marcadores de spin situados tanto na superfície da membrana quanto em seu interior. Considerando um aumento na área por cabeça polar do lipídio após a transição principal, e o modelo de Stern-Gouy-Chapman, foi possível mostrar que a transição causa um aumento no grau de dissociação da vesícula, aumentando portanto a concentração de íons em solução. Entretanto, dentro da estrutura desse modelo simples, foi necessário considerar diferentes constante de associação Na+-DMPG nas fases gel e líquido cristal, para explicar um aumento na repulsão entre as vesículas com a transição principal. Os mecanismos que governam a pós-transição são ainda menos claros.

The temperature dependence of the intensity of light scattered by aqueous dispersions of the anionic lipid DMPG (dimyristoyl phosphatidylglycerol) was studied at different ionic strengths. The lipid main transition, gel-liquid crystal, can be well monitored by a sharp decrease in turbidity. As expected, the temperature of the main transition (Tm) was found to increase with the increase of the ionic strength. For low ionic strength, a DMPG second temperature transition, named post-transition, can be monitored by both an increase in light scattering and a decrease in conductivity. Zimm plot analysis indicates that bellow Tm the liposomes tend to aggregate, yielding a negative second virial coefficient A2, and particles of large molecular weight. At the phase transition, parallel to the decrease in turbidity, there is an increase in the sample conductivity, A2 becomes positive and the particle molecular weight decreases, indicating a non-aggregated state. Moreover, at the post-transition (Tp) A2 becomes very small, and the molecular weight increases again. Both the main and post transitions were found to be reversible. Opposite to the main transition, the post-transition could not be detected by spin labels placed either at the membrane surface, or in the bilayer core. Considering an increase in area per lipid headgroup upon the main transition, and the Stern-Gouy- Chapman model, it was possible to show that the main transition increases the vesicle degree of dissociation, hence increasing the ion concentration in solution. However, within the framework of that simple model, it was necessary to consider different Na+- DMPG association constants, in the gel and liquid phases, to explain an increase in vesicles repulsion upon the main transition. The mechanisms governing the posttransition are even less clear.
