A homeostasia glicêmica requer uma estreita regulação quantitativa e temporal do fluxo de glicose em diferentes órgãos. O fluxo de glicose no fígado e no rim depende de um transportador específico, o GLUT2. Neste estudo demonstramos que em rim, o GLUT2 está aumentado no diabetes e que os fatores transcricionais HNF-1a, HNF-3b e HNF-4a são importantes reguladores do GLUT2, uma vez que apresentaram aumento de expressão e da atividade de ligação no promotor do SLC2A2 (gene do GLUT2). Esses efeitos foram revertidos pelo tratamento com insulina durante 6 dias. No fígado, o diabetes induziu aumento da expressão do GLUT2, HNF-1a, 3b e 4a, assim como aumento na atividade de ligação destes fatores transcricionais no promotor do SLC2A2. Estes resultados evidenciam os HNF- 1a, 3b e 4a como reguladores da expressão do GLUT2 também em fígado de animais diabéticos. No fígado, a insulina reverteu estas alterações aos 4 e 6 dias de tratamento. Os fatores transcricionais SREBP-1c e C/EBP-b não foram alterados em fígado e rim de ratos diabéticos, tratados ou não com insulina.

The glucose homeostasis requires a close temporal and quantitative regulation of the flow of glucose into various organs. The flow of glucose in the liver and kidney depends on a specific transporter, the GLUT2. This study demonstrated that in kidney, GLUT2 is increased in diabetes and that the transcriptional factors HNF-1a , HNF-3b and HNF-4a are important regulators of GLUT2, as it showed increased expression and binding activity in promoter of the SLC2A2 (GLUT2 gene). These effects were reversed by insulin treatment for 6 days. In the liver, diabetes induced increased expression of GLUT2, HNF-1a, 3b, and 4a, as well as increased binding activity of these transcription factors in the promoter of SLC2A2. These results demonstrate the HNF-1a, 3b, and 4a as regulators of the expression of GLUT2 also in liver of diabetic animals. In the liver, insulin reversed these changes at 4 and 6 days of treatment. The transcription factors SREBP-1c and C/EBP-b were not altered in liver and kidney of diabetic rats, treated or not with insulin.
