Para investigar a modulação do estradiol na inflamação gerada no intestino e suas repercussões sistêmicas utilizamos grupos de ratos (Wistar, 60 dias, 160g) submetidos a oclusão da artéria mesentérica superior (45 min), seguida por 2 h de reperfusão. Estudando o efeito de dois protocolos de tratamento com estradiol, terapêutico (30 min após a isquemia - 280 μg/kg, i. v.) ou preventivo (24 h antes da isquemia - 280 μg/kg, s. c.). Como controle utilizamos ratos falsamente submetidos à isquemia (Sham) e ratos não manipulados (Basal). Nossos resultados mostraram que a IR intestinal desencadeia uma inflamação local, isoladamente o intestino e de sistemicamente. Ambos protocolos diminuíram o TNF-α e leucócitos do fluido intestinal, MPO no intestino, LDH e alguns mediadores. Apenas o tratamento preventivo reduziu a permeabilidade vascular intestinal, aumento da motilidade intestinal e a contagem de células da medula óssea. O tratamento terapêutico mostrou diminuição da permeabilidade da mucosa intestinal, leucócitos circulantes, fosfatase alcalina e de mediadores.

To investigate the estradiol modulation of inflammation generated in the intestine and its systemic repercussions we used groups of rats (Wistar, 60 days, 160g) submitted to superior mesenteric artery occlusion (45 min), followed by 2 h of reperfusion. Studying two estradiol treatments protocols effects, therapeutic (30 min after ischemia - 280 μg/kg, i. v.) or preventive (24 h before ischemia - 280 μg/kg, s. c.). As control we utilized rats falsely submitted to ischemia (Sham) and rats not manipulated (Naïve). Our results show that intestinal IR triggers a local inflammation, in the intestine isolated and systemically. Both protocols reduced TNF-α and leukocytes in the intestinal fluid, intestinal MPO, LDH and some mediators (IL-1β, IL-10, VEGF, MIP-1α, MCP-1, GRO/KC e IP-10). Only the preventive treatment reduced of intestinal vascular permeability, increased intestinal motility and bone marrow cell count. The therapeutic treatment showed reduction of intestinal mucosal permeability, circulating leukocytes, alcalin fosfatase and mediators (IL-6, TNF-α e GM-CSF).
