O veneno da Bothrops jararaca (VBj) causa inflamação aguda local. Os mastócitos secretam mediadores que desencadeiam a inflamação por sua ação direta ou pelo recrutamento de células. Este trabalho avalia o papel dos mastócitos na inflamação induzida por VBj em camundongos geneticamente selecionados para alta (AIRmax) ou baixa (AIRmin) resposta inflamatória aguda. Os animais foram tratados com o inibidor de mastócito Cromoglicato de Sódio (CROM) e receberam o VBj, e apresentaram formação de edema similar em resposta ao veneno. Os animais AIRmax apresentaram maior dor que os AIRmin, sendo esta inibida por CROM. O VBj aumentou o número de células peritoneais em AIRmax, bem como o número de macrófagos, e diminuiu o número de mastócitos. O VBj também aumentou neutrófilos e a produção de ROS em AIRmax, efeito inibido por CROM. Os resultados mostram, portanto, que os animais AIRmax apresentam maior dor, migração de neutrófilos e macrófagos e produção de ROS que os AIRmin, e que essas reações são reduzidas quando os mastócitos são inibidos.

Bothrops jararaca venom (BjV) causes acute local inflammation. Mast cells acts secreting mediators that trigger inflammatory events through their direct action or the recruitment of other cells. This study aims to evaluate the role of mast cells in inflammation induced by BjV in mice genetically selected for maximum (AIRmax) or minimum (AIRmin) acute inflammatory response. Animals were treated with mast cell degranulation inhibitor cromolyn (CROM) and received BjV, and showed similar edema formation in response to BjV. AIRmax mice exhibited greatest pain than AIRmin, which was inhibited by CROM. BjV increased peritoneal cells number in AIRmax, as well as the number of macrophages, and diminished the number of mast cells. BjV also increased neutrophils and ROS production in AIRmax, which was inhibited by CROM. These results shows, therefore, that AIRmax mice exhibit more pain, macrophage and neutrophil migration, and ROS production than AIRmin mice, being all these reactions reduced when mast cells are inhibited.
