Nerve growth factor (NGF) is related to pain and increase of Substance P (SP). Non-invasively functional magnetic resonance imaging investigates brain areas. Anti-NGF is a treatment for pain, but not investigated in neuropathic pain. We evaluated the behavioral response and the involvement of NGF and SP in the CCI model after Anti-NGF treatment. We observed increased long-term NGF in the dorsal root ganglion (DRG) and spinal cord, as well as increased SP in DRG at both time and only late in the spinal cord. Treatment with Anti-NGF decreased the nociception of the animals. Anti-NGF reversed high levels of NGF and SP in DRG and spinal cord. There was an increase in Fos in CCI group and a decrease after Anti-NGF treatment in the anterior cingulate cortex. We observed changes in the connectivity of the thalamus, primary somatosensory cortex and cingulate cortex with the entire brain. We hope that these results can be used to develop a therapeutic strategy that will help patients with chronic neuropathic pain.
