A contaminação dos ecossistemas aquáticos por poluentes sintéticos produzidos pelas atividades humanas tem impactado o equilíbrio ambiental global. O crescimento e envelhecimento populacional tem aumentado o consumo de compostos farmacêuticos, como diclofenaco (DCF) e cafeína (CAF), hegemonicamente presentes e persistentes nos ambientes aquáticos. A preocupação emergente de tais compostos se baseia nos efeitos sobre a fisiologia reprodutiva de organismos aquáticos, bem como a ocorrência dos mesmos na forma de misturas. O presente estudo teve por objetivo avaliar os ajustes da fisiologia reprodutiva de machos de Astyanax altiparanae (Teleostei: Characidae) expostos ao DCF e a CAF bem como o potencial sinérgico de DCF+CAF nas respostas fisiológicas dos animais. A. altiparanae é uma importante espécie no contexto ecológico e desenvolvimento social, sendo um dos modelos biológicos mais utilizados em estudos ecotoxicológicos. Inicialmente foi analisado o decaimento (concentração real) de DCF e CAF, que foi de 5% do valor inicial no período de 24 horas e em seguida foi realizada o teste de detecção de alcance, do inglês Range Finding Test (RFT). A concentração média letal (CL50) do DCF e CAF encontrados para o modelo biológico foi de e 30,8 mg.L-1 e 95,9 mg.L-1, respectivamente. No ensaio de exposição aguda (96h, utilizando 10% da CL50), os animais expostos ao DCF (3,08 mg.L-1) e CAF (9,5 mg.L-1) isoladamente, apresentaram uma redução da concentração plasmática de 17β estradiol (E2). Em relação aos andrógenos, os animais apresentaram redução da concentração de testoterona (T) quando expostos ao DCF e não foram observadas diferenças na concentração plasmática de 11 cetotestosternoa (11-KT). No tratamento DCF+CAF não foram observadas diferenças significativas na concentração hormonal de T, E2 e 11-KT. Os fármacos não alteraram a expressão gênica da vitelogenina A (vtgA) e concentração espermática. Os animais também não apresentaram alterações histológicas no fígado e nos testículos durante a exposição aguda. O presente estudo demonstrou que os fármacos isolados afetaram a concentração de esteroides gonadais em machos de A. altiparanae, mas quando expostos em conjunto este efeito não foi observado. O DCF e a CAF tanto isolados quanto misturados em uma exposição aguda não foram capazes de induzir a expressão gênica de vtg, além de não alterar a concentração espermática e modificar a estrutura do fígado e testículos. Considerando a presença de misturas nos ambientes e a importância da reprodução na conservação das espécies, faz-se necessário avaliar os impactos das misturas de fármacos tanto exposições agudas quanto crônicas visto que os efeitos tóxicos dos fármacos, na fisiologia reprodutiva de peixes, podem ser ocultos pelas misturas

The contamination of the aquatic ecosystems caused by anthropogenic pollutants has enhanced the disturbance of the environment balance in a global scale. The overgrowing population and ageing increase the pharmaceuticals consumption, like diclofenac (DCF) and caffeine (CAF) characterized by the ubiquitous presence and persistence in the aquatic environments. The emerging concern behind these compounds is based on the pharmaceuticals mixtures that occur in the environment and the effects of these substances in the reproductive physiology of aquatic organisms. The aim of the present study was to evaluate the physiological reproductive adjustments of Astyanax altiparanae (Teleostei: Characidae) males exposed to DCF and CAF, as well as, the potential synergism of these pharmaceuticals in the physiological responses of the animals. A. altiparanae is considered an important species in the ecological and social development context, also this species is currently used as biological model in ecotoxicological studies. The DCF and CAF decay were 5% of the total initial value (1 mgL-1) in 24 h, then the Range Finding Test (RFT) was carried out. The lethal concentration 50 (LC50) of the DCF and CAF for the species were 95,9 mgL-1 e 30,8 mgL-1, respectively. In the acute exposure (96h, using 10% of LC50), males exhibited a reduction of 17β Estradiol (E2) plasma concentration when exposed to DCF (3,08 mg.L-1) and CAF (9,5 mg.L-1). Similarly, the concentration of testosterone (T) was also reduced under DCF exposure, but this response was not observed in plasma 11 ketotestosterone (11-KT) concentration. Animals exposed to DCF+CAF did not show any difference in T, E2 e 11-KT plasma concentrations. The vitellogenin (vtg) gene expression was also measured but none of the treatments altered significantly the expression, as well as the spermatic concentration. Moreover, the acute exposure did not reveal any morphological alteration in liver and testes. The present study elucidated the effects of single pharmaceuticals in the male steroid profile of A. altiparane and the annul response caused by the mixture. Despite of the absence of interference in the vtg expression, as well as lack of damage within testes and liver, this study showed that pharmaceuticals can impact hormone profile. Considering the presence of mixtures in the environment and the importance of reproduction in species conservation, it is necessary to evaluate the impacts of drug mixtures on both acute and chronic exposures, as the toxic effects of drugs on the reproductive physiology of fish can be hidden by the mixtures
