O tomate (Solanum lycopersicum) é uma das culturas de maior importância econômica atual, além de ser espécie modelo para estudos genéticos em plantas para a qual há diversos recursos genéticos e genômicos bem caracterizados. Tanto o fruto fresco quanto os produtos derivados do tomate constituem fonte de diversos antioxidantes, incluindo a vitamina E (VTE). Em plantas, a síntese dos compostos com atividade de VTE, tocoferóis e tocotrienóis, deriva dos precursores das vias do metileritritol fosfato e do chiquimato. O entendimento dos mecanismos responsáveis pela síntese, transporte e acúmulo da VTE em plantas cultivadas é de grande interesse devido sua implicação para saúde humana e na fisiologia vegetal. Loci para caracteres quantitativos (QTL) para o conteúdo de α-tocoferol em frutos maduros foram previamente mapeados nos cromossomos 6 e 9 em uma população de linhagens introgredidas (ILs) que combinam fragmentos cromossômicos da espécie selvagem Solanum pennellii no fundo genético de S. lycopersicum. Neste trabalho, após busca sistemática em banco de dados de sequências expressas, foram identificados 41 loci envolvidos estruturalmente na biossíntese de VTE em tomate. A análise da distribuição cromossômica desses loci revelou que a maioria dos genes que codificavam enzimas para a rota central do tocoferol estavam localizados nos cromossomos 7, 8 e 9. A variação do conteúdo dos isômeros de tocoferol (α, β, γ e δ) em fruto maduros das ILs foi acessada com intuito de se mapear QTL para cada uma das isoformas. Os perfis metabólicos obtidos permitiram descrever 12 QTL, que corroboram aqueles previamente descritos, além de identificar outros novos nos cromossomos 7 e 8. A análise integrada dos dados metabólicos, genômicos e genéticos levou à proposição de 16 loci candidatos que podem afetar o conteúdo de tocoferol em tomate. A comparação das sequências codificantes desses genes revelou polimorfismos de nucleotídeos e aminoácidos entre os alelos de S. lycopersicum e S. pennellii. Em conjunto, esses resultados representam um importante passo para o entendimento dos determinantes genéticos envolvidos na variação natural do conteúdo de VTE em frutos de tomate.

Tomato (S. lycopersicum) is one of the most important worldwide crops, besides being a relevant model species for plant genetics studies in plants for which there are many genetic and genomic resources well characterized. Both, fresh fruits and tomato-derived products are source of several antioxidants, including vitamin E (VTE). In plants, the synthesis of VTE compounds, tocopherol and tocotrienol, derives from precursors of the shikimate and methylerythritol phosphate pathways. Understanding the mechanisms underlying synthesis, transport and accumulation of VTE in crops are of great interest because of its implications for human health and its role in plant physiology. Quantitative trait loci (QTL) for α-tocopherol content in ripe fruit for chromosome 6 and 9 have been mapped previously in a population of introgression lines (ILs) containing chromosome segments of the wild species Solanum pennellii in the genetic background of the S. lycopersicum. In this work, after a systematic survey in expressed sequences database, 41 loci involved structurally on VTE biosynthesis were identified. The analysis of chromosome distribution of these loci revealed that the majority of genes that encode enzymes for the tocopherol core pathway were located on chromosomes 7, 8 and 9. The variation of tocopherol isomers content (α, β, γ and δ) in ripe fruits of ILs was addressed in order to identify QTL for each isoform. The metabolic profiles obtained allowed to describe 12 QTL that corroborate those previously described while novel ones were identified in chromosome 7 and 8. The integrated analysis at the metabolic, genetic and genomic levels lead to propose 16 candidate loci putatively affecting tocopherol content in tomato. A comparative analysis revealed polymorphisms at nucleotide and amino acid levels between S. lycopersicum and S. pennellii candidate alleles. Together, these results represent an important step for understanding the genetic determinants of VTE natural variation in tomato fruit.
