Pseudomonas aeruginosa é uma bactéria oportunista de ampla distribuição e que infecta principalmente pacientes acometidos por fibrose cística, pneumonia ou com doenças respiratórias pré-existentes, sendo grande responsável por mortes em UTIs em todo o mundo. Entretanto, os mecanismos de ação envolvidos na determinação da virulência desse microrganismo ainda não estão completamente elucidados. A peroxidase Ohr e seu regulador transcricional OhrR, das famílias Ohr/OsmC e MarR (Multiple antibiotic resistance Regulator), respectivamente, estão envolvidos na defesa antioxidante de bactérias a peróxidos orgânicos e peroxinitrito. No presente trabalho foram investigadas as ações de Ohr e OhrR em interações patógeno-hospedeiro por meio de um modelo de infecção pulmonar aguda em camundongos pela linhagem PA14 de P. aeruginosa, bem como pelos mutantes simples (Δohr e ΔohrR) e o duplo mutante (Δohr/ΔohrR). Foram construídas linhagens mutantes apolares por recombinação homóloga e a verificação fenotípica ocorreu por meio dos testes de Kirby-bauer e MIC (Minimal Inhibitory Concentration), confirmando maior sensibilidade das mutantes Δohr e ohr/ΔohrR quando desafiadas por hidroperóxidos orgânicos em relação à selvagem. Inicialmente, as curvas de sobrevivência de camundongos infectados pelas diferentes linhagens foram determinadas. Não houve diferenças estatísticas significativas entre as linhagens, mas os camundongos infectados pela linhagem ΔohrR demonstraram maior capacidade de sobrevivência, sugerindo a participação de OhrR na virulência da bactéria. Ao mesmo tempo, observamos a morte precoce de camundongos infectados pela linhagem Δohr, apontando que a ausência de Ohr poderia estar interferindo de maneira negativa para o hospedeiro. A baixa virulência da linhagem ΔohrR foi um resultado surpreendente, uma vez que a expressão da peroxidase Ohr está aumentada nesses casos. Também foi inesperado o fato da linhagem Δohr provocar mortes precoces. Diversos parâmetros relacionados à resposta inflamatória de camundongos à infecção pelas distintas linhagens de P. aeruginosa foram analisados. Interessantemente, os camundongos infectados pelas linhagens Δohr e Δohr/ΔohrR apresentaram maior recrutamento de neutrófilos de forma significativa em relação à linhagem selvagem. Os níveis de IL-1β e TNF- α nos pulmões de camundongos infectados pela linhagem Δohr foram maiores do que na linhagem selvagem, embora os valores só tenham sido significativos no primeiro caso. Em geral, os níveis de citocinas foram sempre maiores nos pulmões de camundongos infectados com as linhagens mutantes do que com a linhagem selvagem, embora em muitos casos sem significância estatística. O maior recrutamento de neutrófilos e os maiores níveis de citocinas nos pulmões de camundongos infectados pela linhagem Δohr são sugestivos de resposta inflamatória mais intensa. As análises de citocina dos demais órgãos (fígado e baço), bem como a análise de CFU, não apontaram diferenças estatísticas entre as linhagens, o que é coerente com o fato de P. aeruginosa ser um patógeno que infecta pulmões. Uma outra linha de investigação dessa tese envolveu análises bioquímicas e cinéticas de OhrR recombinante de P. aeruginosa (PaOhrR), já que esse fator de transcrição é considerado o sensor bacteriano de peróxidos orgânicos. As reações de PaOhrR com hidroperóxidos orgânicos foram avaliadas por meio da fluorescência intrínseca de triptofano usando fluorímetro acoplado a equipamento de mistura rápida com fluxo interrompido (Stopped Flow). Dessa forma, as reações de PaOhrR com hidroperóxidos derivados de ácidos graxos procedeu com constantes de velocidade na ordem de 106 M-1.s-1. De maneira semelhante, obtivemos valores na ordem de 105 M-1.s-1 para reações com peroxinitrito. Ao mesmo tempo, confirmou-se a baixa reatividade entre PaOhrR e H2O2, com constantes de velocidade na ordem de 101 M-1.s-1. Também construímos mutantes individuais das três cisteínas presentes em PaOhrR, e assim pudemos determinar que Cys19 é a cisteína catalítica e Cys 121 é a cisteína de resolução. Esses resultados foram confirmados por meio de ensaios cinéticos e de SDS-PAGE não redutor. Isso era esperado, já que esses resíduos são conservados entre várias proteínas OhrR. As constantes de velocidade de oxidação de OhrR de C. violaceum (CvOhrR), também investigadas nessa tese, foram bastante semelhantes às constantes de PaOhrR. As constantes cinéticas de oxidação de CvOhrR por hidroperóxidos orgânicos artificiais e derivados de ácidos graxos situaram-se na ordem de 104 M-1.s-1 e 106 M-1.s-1, respectivamente, portanto, valores similares aos obtidos por PaOhrR. Também determinamos a constante cinética de redução de CvOhrR por tiorredoxina de C. violaceum (CvTrxA), resultando em valores da ordem de 104 M-1.s-1. Em geral, as constantes de reação dos fatores de transcrição OhrR estudados nessa tese, estão de duas a três ordens de grandeza menores do que valores correspondentes para Ohr, determinados previamente por nosso grupo. Esses resultados estão de acordo com a proposição de que OhrR teria um papel na percepção de hidroperóxidos orgânicos, enquanto Ohr teria a função de decompor esses oxidantes. Portanto, os resultados obtidos nesta tese contribuíram para o melhor entendimento dos papeis do sistema Ohr-OhrR na resposta bacteriana a peróxidos orgânicos e possivelmente na compreensão de mecanismos de virulência de P. aeruginosa

Kinetic characterization of the transcriptional factor of the Ohr/OhrR system from Pseudomonas aeruginosa and phenotypic and immunologic responses

Pseudomonas aeruginosa is an opportunistic pathogen of widespread distribution that infects especially patients with cystic fibrosis, pneumonia or with pre-existing respiratory diseases, hence being a major cause of death in intensive care units worldwide. Nevertheless, the mechanisms involved in the virulence determination of this microorganism are still elusive. The peroxidase Ohr and its transcriptional regulator OhrR, from Ohr/OsmC and MarR (Multiple antibiotic resistance Regulator) families, respectively, are involved in the antioxidant defense in bacteria against organic hydroperoxides and peroxynitrite. In this research, we investigated the actions of Ohr and OhrR in host-pathogen interactions, using the acute pulmonary infection model in mice with the wild type strain PA14 of P. aeruginosa, as well as with the single mutants (Δohr and ΔohrR) and the double mutant (Δohr/ΔohrR). Nonpolar strains were obtained by homologous recombination and the phenotypic verification was checked by the Kirby-bauer and MIC (Minimal Inhibitory Concentration) tests, confirming higher sensibility of Δohr and ohr/ΔohrR mutants, when challenged by organic hydroperoxides and compared to the wild type (WT) strain. Initially the survival curves of mice infected with the different strains were determined. There were no significant statistical differences among strains, but the mice infected by the ΔohrR strain showed higher survival rates, suggesting the participation of OhrR in the bacterial virulence. At the same time, we observed the early death of mice infected by the Δohr strain showing that the lack of Ohr could be negatively interfering with the host. The low virulence of the δohrR strain was a surprising result as the Ohr peroxidase is higher in these cases. It was also surprising the fact that the Δohr strain causes early deaths. Many parameters related to inflammatory response of mice to the infection by the strains of P. aeruginosa were analyzed. Interestingly enough, the mice infected by the strains Δohr and Δohr/ΔohrR showed significantly higher recruitment of neutrophils compared to the wild type strain. The levels of IL-1β and TNF- α were higher in the lungs of mice infected with Δohr in comparison to the WT strain, although only the first case showed statistical significance. In general, the cytokine levels in the lungs of mice infected with the single and double mutant strains were higher, but in many cases with no statistical significance. The higher neutrophils recruitment and higher levels of cytokines suggested higher inflammatory response caused by the Δohr strain infection. The analysis of cytokines in the other organs (liver and spleen) as well as CFU analysis did not show statistical differences among strains which is coherent with the fact that P. aeruginosa is a lung pathogen. Another field of investigation of this thesis involved biochemical and kinetic analysis of the recombinant OhrR from P. aeruginosa (PaOhrR), as this transcription factor is considered the bacterial sensor for organic hydroperoxides. The reactions of PaOhrR with organic hydroperoxides were evaluated by the intrinsic fluorescence of tryptophan, using a fluorimeter attached to a stopped flow device. The rate constants of the reactions between PaOhrR and fatty acid hydroperoxides were in the order of 106 M-1.s-1, while rate constants for the reaction of PaOhrR with peroxynitrite were within the 105 M-1.s-1 range. In contrast, the reactivity of PaOhrR to H2O2 was very low (in the order of 101 M-1.s-1). We also obtained mutant proteins in which each one of the three cysteines of PaOhrR were individually replaced by Ser residues, which enabled us to determine that Cys19 is the catalytic cysteine and Cys121 is the resolution cysteine. These results were confirmed by kinetic and non-reducing SDS-PAGE assays. These results were expected, as Cys19 and Cys121 residues are conserved across many OhrR proteins. The rate constants for the oxidation of OhrR from C. violaceum (CvOhrR), also investigated here, were very similar to the rate constants determined for PaOhrR. The rate constants for the oxidation of CvOhrR by artificial organic hydroperoxides and fatty acids derivatives were in the order of 104 M-1.s-1 and 106 M-1.s-1, respectively, again similar to the values obtained for PaOhrR. We also determined the rate constant of CvOhrR reduction by thioredoxin from C. violaceum (CvTrxA), resulting in values in the order of 104 M-1.s-1. In general the rate constants of the OhrR transcription factors studied in this thesis are from two to three orders of magnitude lower than the values corresponding to Ohr, previously determined by our research group. These results are in accordance with the proposition that OhrR would have a role in organic hydroperoxides perception while Ohr would be responsible for decomposing these oxidants. Hence, the results obtained in this thesis have contributed to a better understanding of the roles in the Ohr-OhrR system in the bacterial response to organic hydroperoxides and, possibly, in the comprehension of the mechanisms underlying P. aeruginosa virulence
