Abstract in English
The respiratory mechanics of rodents is widely assessed through the impedance of the respiratory system, which might be attained by the forced oscillation technique (FOT), using a mechanical ventilator for small animals (SAV). This method is intrinsically dependent on a physical characteristic of the animal, the body mass, that is used by the SAV to adjust the amplitude of flow perturbations required by the FOT. In order to better understand the relationship between the assessment of the respiratory system mechanics (by FOT) and the physical characteristics of rodents, this work assessed the relationship of the mechanical properties of the respiratory system, using parameters of the constant phase model (MFC) (Rn, G e H), with morphological measurements of male BALB/c mice 18 week-old such as body mass, nose-to-anus length (CNA), Lee index (IL) and ex vivo lung volume. To estimate the total ex vivo lung volume of the animals, it was developed an apparatus that uses the Archimedes's principle. This study was conducted with an animal model of lung inflammation and a control group. The respiratory mechanics was assessed in the basal condition under two ways of injection of phosphate-buffered saline (PBS) via right jugular vein: bolus (B) and continuous infusion (I). Hence, four groups were assessed: OVA-B (n=6), which represents the animal model of lung inflammation with PBS injection via bolus; Controle-B (n=7), which symbolizes the control group of OVA-B; OVA-I (n=8), the animal model of lung inflammation, but with PBS injection via continuous infusion; and, at last, the Controle-I, which indicates the control group of OVA-I. Significant negative correlations were observed in the group Controle-I for the parameters G and H with the body mass (p < 0, 001 and p = 0, 011, respectively) and CNA (p = 0, 001 and p = 0, 001, respectively). We posit that these significant associations observed in the group Controle-I are more related to the method used to assess the respiratory mechanics of these animals (FOT), where the ones with higher value of body mass receive higher amplitudes of flow perturbations, than due to the changes in the volume (size) of the lungs among these animals, because it was not observed any positive and significant correlation of the ex vivo lung volume estimated with body mass or CNA. Thereby, it was also noted that the parameters of the MFC, attained by FOT, are more susceptible to changes due to the volume/flux of air within the lungs, capable of changing functional conditions in central and periphery regions, than due to small anatomical changes in the volume of the lung among mice of the same strain.
