Entre as várias terapêuticas propostas para o tratamento da osteoporose, a administração de altas doses de fluoreto é uma alternativa que ainda apresenta controvérsia na literatura. O fluoreto se destaca por influenciar na atividade dos osteoblastos, os quais são responsáveis por mediar a síntese de moléculas que compõem o osso. No entanto, o mecanismo de ação do fluoreto em células do tecido ósseo (osteoblastos, osteócitos e osteoclastos) ainda não está bem esclarecido. Neste trabalho foram utilizados camundongos com diferentes densidades ósseas, sendo 32 da linhagem C57BL/6J (menor densidade óssea) e 32 da linhagem C3H/HeJ (maior densidade óssea) com 60 dias de vida, os quais receberam água deionizada ou água com 50 ppm de fluoreto (na forma de NaF). Realizou-se os procedimentos cirúrgicos extraindo o incisivo superior esquerdo e posteriormente os animais foram eutanasiados 7, 14, 21 e 28 dias após a extração. Imediatamente após a eutanásia, foi removida a peça com alvéolo dental esquerdo. Foram feitas análises descritiva e morfométrica das lâminas obtidas (coloração em Hematoxilina Eosina). Foi utilizada análise ANOVA a quatro critérios seguida da análise de Tukey (p<0,05). Em geral, para todos os grupos, a análise histológica mostrou neoformação óssea à partir das paredes do alvéolo e formação de ilhotas ósseas invadindo o centro dos alvéolos, substituindo o tecido conjuntivo existente. O tecido ósseo neoformado substituiu aos poucos o tecido conjuntivo ao longo dos períodos experimentais, sendo esse fenômeno ligeiramente mais evidente nos camundongos da linhagem C57BL/6J. A análise morfométrica confirmou os achados histológicos em relação à quantidade de tecido ósseo neoformado e mostrou diminuição do tecido ósseo neoformado no último período (28 dias) nos animais C3H/HeJ (grupos controle e experimental). Concluimos que o processo de reparo ósseo alveolar é semelhante nas duas linhagens estudadas, porém apresentam padrão de formação óssea diferentes entre elas. O fluoreto parece influenciar as etapas do processo de reparo alveolar nas duas linhagens, aumentando a quantidade de osso neoformado, no entanto, a linhagem de camundongos C57BL/6J é mais propensa aos efeitos do flúor.

Among the various therapeutic proposals for the osteoporosis treatment, the administration of high doses of fluoride is an alternative that still has controversies in literature. Fluoride is highlighted by influencing the activity of osteoblasts, which are responsible for mediating the synthesis of bone. But, the mechanism of action of fluoride on bone cells (osteoblasts, osteocytes and osteoclasts) is not well understood. This way, this study aims to evaluate the alveolar bone repair in rats under the effect of fluoride (NaF) administered in the water. This work were used four groups of strains C57BL/6J mice (total of 32) and C3H/HeJ (total of 32) for 60 days, which received deionized water or water with 50 ppm of fluoride. The surgical procedures were made by extracting the upper left incisor and then the animals were euthanized 7, 14, 21 and 28 days. Histological analysis (Hematoxylin Eosin staining), by light microscopy, were made. The statistical analysis were made using the statistical test one-way ANOVA complemented by Tukey's test (p <0.05). Generally, for all groups, the histological analysis showed a new bone formed from the alveolar walls, replacing the connective tissue. The newly formed bone tissue gradually replaces the connective tissue over time; wich was more evident in C57BL/6J mice. Morphometric analisys confirmed the histological findings of the new bone formed and showed a decrease in the new formed bone in the last period (28 days) of the C3H/HeJ animals. We concluded that the process of alveolar repair is similar when it respects to the stages of repair, however they presents different pattern of new bone formation. The fluoride influences the stages of alveolar repair in both lineages, increasing the amount of bone formed, however the mice lineage of C57BL/6J is more prone to the effects of fluoride.
