A dor neuropática é um tipo de dor crônica causada por lesão ou doença no sistema nervoso somatossensorial. A fisiopatologia da dor neuropática é complexa, portanto, fazse necessária uma compreensão dos mecanismos subjacentes para que seja possível identificar novos alvos para intervenção terapêutica. Conforme sugerido por estudos recentes, modificações pós-traducionais em proteínas-alvo do sistema somatossensorial podem contribuir para a fisiopatologia de diversas doenças. A O-GlcNacilação induzida pela N-AcetilGlucosamina Transferase (OGT), um tipo de modificação pós-traducional, promove a ligação reversível de N-acetilglucosamina, produto final da via das hexosaminas, em resíduos de serina e treonina de proteínas. Apesar de a O-GlcNacilação ter uma importância biológica na modulação da função proteica em diversos contextos fisiopatológicos, seu papel na fisiopatologia da dor neuropática permanece desconhecido. Assim sendo, o objetivo deste trabalho foi investigar o envolvimento da O-GlcNacilação no desenvolvimento da dor neuropática. A avaliação da expressão gênica e proteica de enzimas pertencentes à via das hexosaminas e da atividade de OGT na medula espinal de comundongos C57/BL6, submetidos ao modelo de dor neuropática por injúria de nervos periféricos com nervo poupado (do inglês spared nerve injury - SNI), revelou um aumento significativo. Foi observado também que a expressão de OGT na medula espinhal estava confinada quase que exclusivamente em astrócitos. Por fim, foi verificado que a ausência de OGT em astrócitos reduz o desenvolvimento da dor neuropática no modelo de SNI. Este estudo demonstra pela primeira vez um papel funcional da O-GlcNacilação para o desenvolvimento da dor neuropática, apontando para um possível alvo para o desenvolvimento de novas abordagens terapêuticas

Neuropathic pain is a type of chronic pain caused by injury or disease in the somatosensory nervous system. The pathophysiology of neuropathic pain is complex, therefore, it is necessary to understand the underlying mechanisms to be possible identify new targets for therapeutic intervention. As suggested by recent studies, post-translational modifications in target proteins of the somatosensory system may contribute to the pathophysiology of various diseases. O-GlcNacylation induced by N-Acetyl Glucosamine Transferase (OGT), a type of post-translational modification, promotes the reversible attachment of N-acetylglucosamine, the end product of the hexosamine pathway, to serine and threonine protein residues. Although O-GlcNacylation has biological importance in the modulation of protein function in various pathophysiological contexts, its role in the pathophysiology of neuropathic pain remains unknown. Therefore, the objective of this work was to investigate the involvement of O-GlcNacilation in the development of neuropathic pain. The evaluation of the gene and protein expression of enzymes belonging to the hexosamine pathway and the OGT activity in the spinal cord of C57 /BL6 mice submitted to the neuropathy model of spared nerve injury (SNI), showed a significant increase. It was also observed that OGT expression in the spinal cord was confined almost exclusively in astrocytes. Finally, it was found that the absence of OGT in astrocytes reduces the development of neuropathic pain in the SNI model. This study demonstrates for the first time a functional role of O-GlcNacilation for the development of neuropathic pain, pointing to a possible target for the development of new therapeutic approaches
