A regulação do balanço energético envolve interação de diferentes fatores metabólicos que sinalizam para o sistema nervoso o status energético do organismo. O hipotálamo integra esses sinais de forma coordenada por meio de circuitos neuronais envolvendo diferentes núcleos interconectados, como o núcleo arqueado (ARC), o núcleo paraventricular (PVN), o núcleo dorsomedial (DMH), núcleo hipotalâmico ventromedial (VMH) e a área hipotalâmica lateral (LHA), que regulam o apetite, a saciedade, o gasto energético e a homeostase da glicose. No VMH, há os neurônios que expressam o fator esteroidogênico 1 (SF1), que participa de muitos processos fisiológicos, incluindo a homeostase energética. Relacionado ao balanço energético, a leptina tem ação fundamental reduzindo a ingestão alimentar e aumentando o gasto de energia. A leptina age no hipotálamo por meio da ligação ao seu receptor, LepR, e promove a fosforilação e ativação da tirosina quinase janus-kinase-2 (JAK2) que, por sua vez, fosforila fatores de transcrição chamados de STATs (transdutor de sinal e ativador da transcrição), como a STAT5, que regulam a expressão dos genes-alvo da leptina. Outros fatores promovem também a ativação de STAT5 em neurônios hipotalâmicos, como o fator estimulador de colônias de granulócitos e macrófagos (GM-CSF), uma citocina pró-inflamatória. Alguns trabalhos demonstraram que a injeção central de GM-CSF no terceiro ventrículo cerebral diminuiu a ingestão alimentar e o peso corporal em ratos e camundongos, sendo que essa resposta é atenuada em animais com deleção de STAT5 no sistema nervoso central (SNC), indicando um papel da via da STAT5 no SNC na regulação da ingestão de alimentos por meio da ativação pelo GM-CSF. Contudo, a participação da via de sinalização da STAT5 em neurônios SF1 do VMH na homeostase energética ainda não está bem estabelecida. Dessa forma, os objetivos deste trabalho foram investigar a importância da via de sinalização da STAT5 em neurônios SF1 do VMH na homeostase energética, assim como avaliar a participação desta via de sinalização nos efeitos hipofágicos promovidos pelo GM-CSF. Para tanto, utilizamos a metodologia cre-loxP para geração de camundongos com deleção específica de STAT5 em neurônios SF1. Assim, camundongos SF1-cre(cre/+) foram acasalados com camundongos STAT5flox/flox, para obtenção de animais SF1-cre(cre/+); STAT5flox/flox, com deleção de STAT5 especificamente em neurônios SF1, e animais controles STAT5flox/flox. Foram avaliados peso corporal, ingestão alimentar, gasto energético, metabolismo glicêmico e concentrações séricas de insulina e leptina, em animais experimentais e controles, machos e fêmeas, tratados com dieta normocalórica ou hiperlipídica. Avaliamos também o peso corporal e a ingestão alimentar dos camundongos SF1-cre(cre/+); STAT5flox/flox e STAT5flox/flox machos, tratados com dieta normocalórica e após receberem dose única de GM-CSF (0,5 µg/2 µl) ou veículo (2 µl) intracerebroventricular. Observamos que o peso corporal, a ingestão de alimentos, o gasto energético, o metabolismo glicêmico e os níveis séricos de leptina não se diferiram entre os grupos de animais experimentais SF1-cre(cre/+); STAT5flox/flox e seus controles STAT5flox/flox, tanto machos como fêmeas, com dieta regular ou hiperlipídica. Em relação aos parâmetros metabólicos avaliados nos animais machos, após receberem o estímulo com GM-CSF icv, observamos que a citocina induziu a redução da ingestão alimentar somente entre os animais controles STAT5flox/flox. A redução do peso corporal foi maior e mais persistente no grupo de animais controles STAT5flox/flox, quando comparados aos animais experimentais SF1-cre(cre/+); STAT5flox/flox. Em conclusão, a deleção da via de sinalização da STAT5 em neurônios SF1 do VMH não afeta a homeostase energética em condições de dieta regular ou obesogênica. Por outro lado, esta via de sinalização participa nas ações do GM-CSF sobre a ingestão alimentar e o peso corporal. A participação da via de sinalização da STAT5 em neurônios SF1 do VMH nos efeitos catabólicos de outras citocinas requer investigação futura.

The regulation of the energy balance involves interaction of different metabolic factors that signal to the nervous system the energy status of the organism. The hypothalamus integrates these signals in a coordinated way through neuronal circuits involving different interconnected nuclei, such as the arcuate nucleus (ARC), the paraventricular nucleus (PVN), the dorsomedial nucleus (DMH), the hypothalamic ventromedial nucleus (VMH) and the lateral hypothalamic area (LHA), which regulate appetite, satiety, energy expenditure and glucose homeostasis. In VMH, there are neurons that express steroidogenic factor 1 (SF1), which participates in many physiological processes, including energetic homeostasis. Related to the energy balance, leptin has a fundamental action reducing food intake and increasing energy expenditure. Leptin acts on the hypothalamus through binding to its receptor, LepR, and promotes the phosphorylation and activation of the janus-kinase-2 tyrosine kinase (JAK2), which in turn phosphorylates transcription factors called STATs (signal transducer and transcriptional activator), such as STAT5, that regulate expression of the target genes of leptin. Other factors also promote the activation of STAT5 in hypothalamic neurons, such as the granulocyte-macrophage colony-stimulating factor (GM-CSF), a proinflammatory cytokine. Some studies have shown that the central injection of GM-CSF in the third cerebral ventricle decreased food intake and body weight in rats and mice, and this response is attenuated in animals with STAT5 deletion in the central nervous system (CNS), indicating a role of the STAT5 pathway in the CNS in regulating food intake through GM-CSF activation. However, the participation of the STAT5 signaling pathway in VMH SF1 neurons in energetic homeostasis is still not well established. Thus, the objectives of this work were to investigate the importance of the STAT5 signaling pathway in VMH SF1 neurons in energetic homeostasis, as well as to evaluate the participation of this signaling pathway in the hypophagic effects promoted by GM-CSF. For this, we used the cre-loxP methodology for the generation of mice with specific deletion of STAT5 in SF1 neurons. Thus, SF1-cre(cre/+) mice were mated with STAT5flox/flox mice, to obtain SF1-cre(cre/+); STAT5flox/flox animals, with STAT5 deletion specifically on SF1 neurons, and STAT5flox/flox control animals. Body weight, food intake, energy expenditure, glycemic metabolism and serum insulin and leptin concentrations were evaluated in experimental and control animals, male and female, treated with normocaloric or hyperlipidic diet. We also evaluated body weight and food intake of SF1-cre(cre/+); STAT5flox/flox and STAT5flox/flox mice, treated with normocaloric diet and after receiving a single dose of GM-CSF (0.5 µg/2 µl) or intracerebroventricular vehicle (2 µl). We observed that body weight, food intake, energy expenditure, glycemic metabolism and serum leptin levels did not differ between groups of experimental animals SF1-cre(cre/+); STAT5flox/flox and its controls STAT5flox/flox , both male and female, on a regular or hyperlipidic diet. Regarding the metabolic parameters evaluated in the male animals, after receiving the stimulation with GM-CSF icv, we observed that the cytokine induced the reduction of food intake only among STAT5flox/flox control animals. The reduction of body weight was greater and more persistent in the group of STAT5flox/flox control animals, when compared to experimental animals SF1-cre(cre/+); STAT5flox/flox . In conclusion, the deletion of the STAT5 signaling pathway in VMH SF1 neurons does not affect energy homeostasis under regular or hyperlipidic dietary conditions. On the other hand, this signaling pathway participates in GM-CSF actions on food intake and body weight. Participation of the STAT5 signaling pathway in VMH SF1 neurons in the catabolic effects of other cytokines requires further investigation.
