Microorganisms in the rhizosphere can operate directly for the benefit of the plant through functions such as defense, health and growth promotion, and the use of these microorganisms as inoculants represents a viable alternative for a more sustainable and balanced agricultural production without achieved the levels of production and technology already in place. In this study, 71 bacteria from the rhizosphere of modern cultivars and wheat landraces were isolated and characterized. The isolates were characterized by sequencing the 16S rRNA gene and selected after biochemical tests for acetic acid-indole production, phosphate solubilization, nitrogen fixation and antagonism against soil borne fungal pathogens. We assessed the contribution of these selected microorganisms for wheat growth promotion in bioassays and we evaluated the impact of inoculation in the rhizosphere bacterial community. The effect of the isolates on the plant was verified in a greenhouse experiment, with 11 treatments, using three commercial wheat genotypes that were evaluated for height, diameter, amount of chlorophyll, dry and fresh weight of the root and aerial part and analysis of the enzymes FDA, β-glycosidase and phosphatase to measure microbial activity in the soil. In addition to seed treatment, biweekly inoculations and rhizospheric soil sampling were carried out throughout the experiment, before and after each bacterial inoculation. It was possible to observe a plant growth promotion especially in plants inoculated with bacteria of the genus Lysinibacillus and Pseudomonas in the cultivar BRS Parrudo, and in the cultivar BRS Reponte the treatment with the bacterial consortium including all isolates tested in the bioassay was the most efficient, indicating that the beneficial interaction is cultivar-dependent. In addition, the permutational multivariate analysis of the 16S rRNA data in soil samples indicated that there is a significant change in the structure of the bacterial community of the rhizosphere over plant development, which is also impacted by the bacterial inoculation. The inoculant is more easily established in the early stages of the plant development and its relative abundance gradually decreases days after inoculation. The results provide conclusions about the influence of the genotype on the plant's response and suggest that the development of inoculants must be cultivar-specific.
