No Brasil, os enfezamentos vermelho e pálido são, presentemente, as principais doenças da cultura do milho, por causarem danos significativos na produção de grãos e sementes. A intensidade de ocorrência destas doenças se agravou a partir da década de 1980, com a implementação dos plantios da segunda safra do milho, também conhecida por "safrinha". Nos últimos anos, grandes surtos da doença têm sido relatados nas principais regiões produtoras do país, mostrando que o patossistema ainda exige muito empenho da pesquisa, além do conhecimento até agora gerado. Buscando contribuir para melhor entendimento das relações entre o hospedeiro e os patógenos envolvidos com ambas as formas de enfezamentos, foi conduzido o presente estudo, com os seguintes objetivos: i- verificar a incidência isolada e simultânea do fitoplasma e do espiroplasma em diversas localidades situadas em distintas regiões brasileiras; ii- analisar a dinâmica populacional de ambos os patógenos na colonização do hospedeiro e sua relação com a expressão de sintomas; iii- avaliar a diversidade do fitoplasma, utilizando o promissor gene marcador denominado SecY. Para análises da incidência dos patógenos, foram realizadas coletas de amostras de plantas sintomáticas em diferentes regiões geográficas brasileiras e as mesmas foram submetidas a reações de PCR multiplex. Resultados mostraram maior frequência de ocorrência do espiroplasma em relação ao fitoplasma, nas áreas geográficas mais quentes. Em condições de campo, foi evidenciado um relevante aumento na incidência de plantas doentes causada pela infecção simultânea de ambos os patógenos, em comparação com relatos feitos em anos anteriores. No ensaio realizado com inoculação simultânea do fitoplasma e espiroplasma por meio do inseto vetor D. maidis e posterior análise com PCR em tempo real, foi possível observar que o espiroplasma ocorreu em maior concentração nas plantas inoculadas simultaneamente por estes patógenos. Com base na expressão dos sintomas, foi observado que, embora tenha ocorrido predominância do espiroplasma, a maioria das plantas experimentalmente submetidas à infecção mista apresentaram avermelhamento foliar, um sintoma típico de infecção por fitoplasma. A caracterização molecular do fitoplasma do milho realizada a partir de clonagem, sequenciamento, análise virtual de RFLP e análise filogenética revelou ausência de diversidade genética do fitoplasma encontrado nas amostras coletadas, em relação à análise do gene 16S rRNA, sendo este fitoplasma classificado como afiliado ao subgrupo 16SrI-B. Análises conduzidas com o gene SecY, permitiram identificar maior variabilidade, sendo o fitoplasma classificado como pertencente ao subgrupo secY(I)-L. O presente trabalho traz sua colaboração no sentido de melhor entendimento sobre este patossistema, sendo que seus resultados atualizam aspectos relacionados à incidência isolada ou simultânea destes patógenos no campo, à colonização do hospedeiro e à identificação molecular baseada em um novo gene marcador.

In Brazil, maize bushy stunt and corn stunt are currently the main diseases of the corn crop. These diseases cause significant damage to grain and seed production. The intensity of occurrence of these diseases worsened from the 1980s, with the implementation of the second crop of corn, also known as "safrinha". In recent years, major outbreaks of the disease have been reported in the main producing regions of the country, showing that the pathosystem still requires a lot of research effort, besides the knowledge generated so far. Aiming to contribute to a better understanding of the relationship between the host and the pathogens that cause these diseases, the present study was conducted with the following objectives: i- to evaluate the isolated and simultaneous incidence of phytoplasma and spiroplasma in several localities situated in different Brazilian regions; ii- analyze the population dynamics of both pathogens in host colonization and their relationship with symptom expression; iii- evaluate phytoplasma diversity using the promising marker gene called SecY. Evaluation of the incidence of pathogens was performed with samples of symptomatic plants collected in different Brazilian geographic regions and submitted to multiplex PCR. Results showed a higher frequency of occurrence of spiroplasma in relation to phytoplasma in warmer geographical areas. Under field conditions, a significant increase in the incidence of diseased plants caused by the simultaneous infection of both pathogens was evidenced compared to reports made in previous years. In the assay performed with simultaneous inoculation of phytoplasma and spiroplasma by D. maidis and subsequent analysis with real-time PCR, it was observed that the spiroplasma occurred in higher concentration in plants simultaneously inoculated by these pathogens. Based on the expression of symptoms, it was observed that although there was predominance of spiroplasma, most plants experimentally submitted to mixed infection presented leaf reddening, a typical symptom of phytoplasma infection. Molecular characterization of phytoplasma by cloning, sequencing, virtual RFLP analysis, and phylogenetic analysis revealed lack of genetic diversity of phytoplasma found in the collected samples in relation to the 16S rRNA gene analysis, which is classified in the subgroup 16SrI-B. Analyzes conducted with the SecY gene allowed to identify greater variability, and the phytoplasma was classified as belonging to the secY(I)-L subgroup. The present work contributes to a better understanding of this pathosystem, and its results update aspects related to the isolated or simultaneous incidence of these pathogens in the field, host colonization and molecular identification based on a new marker gene.
