Para estudar o papel dos rins e o de tratamentos alternativos na desintoxicação da amônia quatro trabalhos foram realizados em ratos e bovinos. Ratos Wistar (n=367) foram utilizados para testar oito tratamentos (O= aminoácidos do ciclo da uréia; F= furosemida; H= solução hidratante, O+F; H+F, O+H; O+F+H, C= controle) no combate à intoxicação por amônia. Acetato de amônio foi injetado pela via intraperitoneal e 3min após os tratamentos eram injetados pela mesma via. Maiores sobrevivências e menores teores de amônia plasmática foram obtidos nos tratamentos O+F+H (62,5%) e O+H (57%). A medicação com O, H e F promoveu maior teor sérico de uréia e menor de creatinina e menos pronunciado edema pulmonar, respectivamente. No segundo trabalho, desenvolveu-se, em 15 garrotes, um modelo de intoxicação por amônia por meio de infusão de cloreto de amônio (NH4+Cl) [iv] até o surgimento de convulsão. O quadro clínico exibido foi idêntico à intoxicação natural e todos os animais sobreviveram. Durante a infusão elevaram-se os teores sangüíneos de amônia, uréia, lactato-L, glicose, potássio e hematócrito, e diminuíram o pH e o bicarbonato sangüíneos. A acidose metabólica ocorreu por aumento do teor de lactato-L; quanto menor o pH do sangue maior o teor de potássio. O modelo de indução foi seguro, prático, de alta reprodutibilidade e não causou efeitos colaterais. Nestes mesmos animais, durante a indução foram estudados o desempenho renal na excreção de amônio e a relação desta com a resistência dos bovinos à intoxicação. Quanto maior o volume de urina excretado durante a infusão maior a quantidade de NH4+Cl injetado para provocar convulsão, assim como maior a quantidade de amônio excretado na urina. Quanto menor o pH sanguíneo e urinário maior o índice de excreção urinário de amônio. Alguns animais ficaram desidratados, mas esta não foi causada pela diurese. Concluiu-se que os rins têm um papel importante na excreção de amônia em bovinos intoxicados e quanto maior a diurese menos propenso o animal é a intoxicação. Em seguida, os bovinos intoxicados foram distribuídos ao acaso e tratados com O+F+H, O+H ou C no decorrer de 3h após o término da infusão de NH4+Cl. Os dois primeiros tratamentos promoveram melhora clínica pronunciada, em relação ao grupo C, diminuíram os teores de amônia e lactato-L, urinaram e excretaram NH4+ mais abundantemente e tiveram menor pH urinário. Os tratamentos propostos foram efetivos na desintoxicação por amônia.

To study the role of the kidneys and of alternative treatments for detoxifying ammonia four experiments were carried out in cattle and rats. Wistar rats (n=367) were used to compare eight treatments (O= urea-cycle amino acids; F= furosemide; H= saline solution, O+F; H+F; O+H; O+F+H; C=control) to reverse ammonia poisoning. Ammonium acetate solution was injected [ip] and 3 min later the treatments were injected through the same via. Higher survival rate and lower plasma ammonia levels were detected in rats treated with O+F+H (62.5%) and O+H (57%). Rats treated with O, H and F had higher urea serum, lower creatinine and less pronounced lung edema, respectively. To develop a practical model to induce ammonia poisoning through [iv] ammonium chloride infusion until the appearance of convulsion (NH4+Cl) 15 yearling steer were used. The clinical picture showed was identical to the natural poisoning. Higher blood levels of ammonia, urea, L-lactate, glucose, potassium and hematocrit and lower bicarbonate and blood pH were detected during the infusion. A metabolic acidosis occurred by the increase of L-lactate; the lower the blood pH, the higher the hipercalemia. The model was very practical, reliable, and highly reproducible; no side effects were seen. All steers survived after the poisoning. During the same trial the role of the kidney to excrete ammonium was studied as well as the relation of this excretion with the individual resistance to the poisoning. The higher the urine volume excreted during the infusion, the higher the volume of NH4+Cl infused to cause convulsion and the greater the amount of NH4+ excreted in the urine. The lower the blood and urine pH, the higher the urinary ammonium excretion rate. Some steers were dehydrated, but dehydration was not cause by diuresis. In conclusion, the kidneys have an important role in the excretion of ammonium in the ammonia poisoning cattle, the higher the diuresis throughout the induction, the more resistant the cattle is to the poisoning. Afterwards, the same poisoned steers were randomly distributed and treated for 3h with O+F+H, O+H or C (small amount of saline - iv). The two former treatments produced a pronounced clinical improvement, lowering the blood ammonium and L-lactate levels and urine pH and increasing the urinary volume and NH4+ excretion. The selected treatments were very effective for detoxifying ammonia in poisoned cattle.
