Em bovinos, a mortalidade embrionária associada a falhas no processo de reconhecimento materno da prenhez atinge 30 a 40%. O sucesso da prenhez , depende de uma apropriada interação bioquímica entre o endométrio materno e o concepto. Objetivou-se a validação de uma técnica cirúrgica de implantação de cateteres intrauterinos para a obtenção de fluídos em diferentes dias do ciclo estral e da prenhez inicial em bovinos, de modo a permitir a análise sistemática e contínua das secreções do microambiente uterino. O funcionamento dos cateteres foi testado e alterações morfológicas conseqüentes da implantação dos cateteres foram descritas. Cateteres de silicone foram inseridos cirurgicamente nos cornos uterinos e exteriorizados pelo flanco de nove vacas Holandesas. As vacas foram divididas para receberem (n = 6) ou não (n = 3) embriões no dia 7 de um ciclo estral sincronizado (dia 0 = dia do cio). Microlavagens uterinas foram realizadas nos dias 14, 16, 18 e 20 pós-estro. Todas as vacas foram abatidas no dia 20, seus tratos reprodutivos submetidos à análise macroscópica e tecidos uterinos coletados e fixados para análise histopatológica. A técnica utilizada: (1) permitiu a obtenção de fluídos uterinos, (2) resultou em 0% de taxa de prenhez, (3) causou afecções nos órgãos reprodutivos, como aderências, infecções focais e endometrites e (4) afetou o tecido uterino com inflamações agudas e crônicas além de fibroplasias e dilatações glandulares. Conclui-se que a implantação e operação dos cateteres e as alterações morfológicas foram incompatíveis com a manutenção da prenhez. A abordagem cirúrgica testada não foi adequada para estudar o microambiente uterino de vacas prenhez prenhes

In cattle, embryonic death related to failure in the maternal recognition of pregnancy is as high as 30 to 40%. The success of pregnancy depends on appropriated biochemical interactions between the maternal endometrium and conceptus. The purposed of this dissertation was to validate a surgical technique to place and operate of intrauterine catheters to obtain fluid on different days of the estrous cycle and early pregnancy in cows, such technique should allow a systematic and continuous analysis of the uterine microenvironment secretions. Function of catheters was tested and resulting morphological changes were described. Silicon indwelling catheters were surgically placed in both uterine horns and exteriorized by the flank in 9 Holstein cows. Cows were divided to either receive (n = 6) or not (n = 3) embryo transfer on the day 7 of a synchronized estrous cycle (day 0 = day of estrus). Uterine flushing was performed through the catheters on days 14, 16, 18 and 20 pos-estrus. All cows were slaughtered on day 20 and reproductive tracts were colleted and prepared for histological analysis. In summary, the technique used: (1) allowed collection of uterine fluids, (2) resulted in 0% pregnancy rate, (3) caused pathologies in the reproductive organ, such as adhesions, focal infections and endometritis and (4) affected the uterine tissue with acute and chronic inflammation besides fibroplasias and glandular dilation. It was concluded that implantation and operation catheters the resulted in morphological changes which were incompatible with maintenance of pregnancy. The surgical approach tested was not suitable to study the uterine microenvironment in pregnant cows
