Sabendo do impacto que nutrição e ambiente podem ter sobre a reprodução é de fundamental importância para aplicação nas biotecnologias da reprodução analisar o ciclo reprodutivo de novilhas bovinas (taurinas e zebuínas) e bubalinas, temporalmente, sob as mesmas condições nutricionais e de manejo. Assim, este estudo teve como objetivo analisar temporalmente a dinâmica folicular de novilhas Holandesas, Gir e Búfalas, mantidas sob as mesmas condições de manejo. As fêmeas foram alocadas no Departamento de Reprodução Animal (CBRA/FMVZ/USP), Campus Pirassununga. No Experimento 1 as fêmeas foram pré-sincronizadas com duas doses de prostaglandina F2α (PGF; Dcloprostenol, 150 µg IM, Prolise®), com 14 dias de intervalo. O dia da primeira ovulação, após segunda dose de PGF, foi considerado como o dia 0 do ciclo. A partir do D0 foram realizadas avaliações diárias da dinâmica folicular (folículos > 3 mm) por ultrassonografia (Aloka SSD500, 7,5MHz), até à detecção da ovulação subsequente. O Experimento 2 foi realizado após a segunda ovulação, avaliando-se a dinâmica folicular a cada 12h para detectar o momento do desvio da primeira onda folicular. As variáveis resposta foram analisadas pelo PROC GLIMMIX do SAS. Não foi encontrada diferença entre os intervalos inter-ovulatórios (22,3±0,65; 22,1±0,95; 23,8±0,63 dias; Holandesas, Gir e Búfalas, respectivamente). No entanto, em relação às características a seguir, foram encontradas diferenças significativas (P<0,05) em pelo menos um dos grupos genéticos, Holandesas, Gir e Búfalas, respectivamente, para o número de ondas foliculares em um ciclo estral (2,6±0,17a, 3,4±0,24b, 2,8±0,11a ondas), número de folículos recrutados após primeira onda (26,1±1,7a, 66,2±15,9b, 22,7±1,91a folículos), e segunda ovulação (30,2±3,08a, 74,2±18,8b, 27,1±2,8a folículos), diâmetro do FD da primeira onda (15,8±0,66a, 12,3±0,32b, 12,1±0,03b mm), diâmetro do FD da segunda onda (11,8±0,73a, 9,4±0,52b, 10,6±0,31ab mm), diâmetro do folículo ovulatório (15±0,33a, 13,7±0,74ab, 13,5±0,46b mm), diâmetro máximo do CL (26,8±0,54a, 22,3±0,98b, 19,5±0,32c mm), concentração máxima de P4 no ciclo (7,7±0,33a, 8,0±0,85a, 5,3±0,28b ng/ml) e duração da dominância do FD da primeira onda (8,0±0,49a, 5,2±1,07b, 8,1±0,44a dias). Ainda com relação aos esteroides, a análise de testosterona plasmática nos 13 primeiros dias do ciclo foi semelhante entre as bovinas e menor para as bubalinas. Insulina e IGF plasmático também foram mensuradas. Novilhas taurinas e bubalinas (4,1±0,67 mUI/ml, 6,0±0,52 mUI/ml, respectivamente) apresentaram menores concentrações de insulina do que as zebuínas (9,0±2,15 mUI/ml), sendo que estas concentrações foram maiores durante a fase luteínica do que na fase folicular. Em relação ao desvio folicular no Experimento 2, não foram encontradas diferenças entre as raças para o momento do desvio (1,8±0,15; 2,1±0,21; 1,8±0,18 dias), entretanto, novilhas Holandesas apresentaram neste momento maior diâmetro de FD e FS (FD: 7,9±0,33; FS: 7,0±0,27 mm; P<0,05) quando comparadas às Gir (FD: 7,0±0,32; FS: 6,2±0,28mm) e às Búfalas (FD: 6,8±0,19; FS: 6,3±0,29mm). Não foram encontradas diferenças para concentrações de IGF-I total no fluído folicular e no plasma das taurinas e das zebuínas, porém as concentrações nas bubalinas foram superiores a das bovinas. Com esses resultados, conclui-se que novilhas de grupos genéticos diferentes (Bos taurus, Bos indicus e Bubalus bubalis) apresentam características de crescimento folicular e luteínico específicos para a raça, quando avaliadas temporalmente e nas mesmas condições de manejo ambiental e nutricional.

Considering the impact that nutrition and environment have over reproduction, it is essential for the reproductive biotechnologies to analyze the reproductive cycle of bovine (taurine and zebu) and bubaline heifers at the same time and under the same nutritional management. Therefore, the objective of this work was to study the follicular dynamic of Holstein (n=15), Gir (n=11) and Buffalo (n=15) heifers kept under the same conditions. All heifers were housed at the Department of Animal Reproduciton, (CBRA/FMVZ/USP), Pirassununga Campus. Animals were pre-synchronized with two doses of prostaglandin F2α (PGF; Dcloprostenol, 150µg, IM), fourteen days apart (D0 = day of first ovulation). From D0, it was performed daily ultrasound scan of the ovaries to track follicular dynamics (follicles ≥ 3mm) until the occurrence of the second ovulation. After the second ovulation, the follicular dynamic was performed twice daily to detect the time of deviation of the first follicular wave. The response variables were analyzed by PROC GLIMMIX, SAS. No difference was found among the inter-ovulatory interval (22.3±0.65; 22.1±0.95; 23.8±0.63 days) for Holstein, Gir and Buffalo, respectively. Differences were present (P<0.05) in at least one of the genetic groups, Holstein, Gir and Buffalo, respectively, for number of follicular waves (2.6±0,17a, 3.4±0,24b, 2.8±0,11a waves), number of follicles recruited after first (26.1±1.7a, 66.2±15.9b, 22.7±1.91a follicles), and second ovulation (30.2±3.08a, 74.2±18.8b, 27.1±2.8a follicles), diameter of first wave DF (15.8±0.66a, 12.3±0.32b, 12.1±0.03b mm), diameter of second wave DF (11.8±0.73a, 9.4±0.52b, 10.6±0.31ab mm), diameter of ovulatory follicle (15±0.33a, 13.7±0.74ab, 13.5±0.46b mm), maximum diameter of CL (26.8±0,54a, 22.3±0.98b, 19.5±0.32c mm), maximum concentration of P4 during ovulatory cycle (7.7±0.33a, 8.0±0.85a, 5.3±0.28b ng/ml) and length (8.0±0.49a, 5.2±1.07b, 8.1±0.44a days) of first wave dominant follicle dominance. No differences were found among the breeds for the moment of deviation (1.8±0.15; 2.1±0.21; 1.8±0.18 days). However, Holstein heifers presented larger DF and SF diameter (FD: 7.9±0.33; FS: 7.0±0.27mm; P<0.05) at deviation when compared with Gir (DF: 7.0±0.32; SF: 6.2±0.28mm) and Buffalo (DF: 6.8±0.19; SF: 6.3±0.29mm). Analyzing other steroids, concentration of blood testosterone during the first 13 days of the cycle was similar for the bovine heifers and lower for the bubaline heifers. Blood insulin and IGF-I were also measured. Holstein and bubaline heifers shown smaller concentrations of insulin (4.1±0.67 mUI/ml, 6.0±0.52 mUI/ml, respectively) than Gir heifers (9.0±2.15 mUI/ml), and also, when grouping the data collected from luteal phase and follicular phase, the result shows that during the period of high P4, insulin concentrations are lower than during the period of high E2. No differences were found among bovine breeds for the IGF-I concentration in follicular fluid, but they were lower than the concentrations presented in the bubaline follicular fluid. The results herein presented show that heifers from different genetic groups (Bos taurus, Bos indicus and Bubalus bubalis), maintained at the same nutritional and environmental status, have different folliculogenesis and luteal growth, which are specific to its breed.
